In recent years,biomedical research have progressed from studies of mono-layers of cells on glass to in situ studies of both ex vivo and in vivo biological system to extract more realistic biological information and therefore,this trend have prompted the development of three-dimensional(3-D)mesoscopic(mm-cm)optical imaging technology.Optical Projection Tomography(OPT)and Scanning Laser Optical Tomography(SLOT),which is an exciting new technique for imaging mesoscopic samples at a high resolution,provides 3-D absorption and/or fluorescence distributions of chromophores in optically transparent specimens including fixed and chemically cleared tissues(e.g.mouse embryo)and inherently transparent live animal models(e.g.zebrafish embryo)and then for biomedical research such as histopathology.It provides a new imaging model for the life sciences,materials science,biomedical research.LabVIEW is an industry-standard graphics software platform,which combines the flexibility in graphical programming and high performance capabilities in test,measurement and automation control.It provides a robust development tool for data acquisition,instrument control,measurement analysis and data display.This paper describes the design of OPT and SLOT based on LabVIEW and exaplores its capabilities to image intact biological organs or tissues.The main contents are as follows:Firstly,investigating the optical imaging principle of OPT and SLOT,both of which are optical CT imaging techniques based on the theory of computed tomography reconstruction,not only have high plane resolution but also have high dept resolution,they can provide two imaging modes,especially SLOT imaging system has a high photon collection efficiency,and compare with other imaging modalities in system constitute,sample processing and data processing.Secondly,introducing the hardware systems and software systems of OPT and SLOT,hardware system consist of light road module,sample placed module,signal detection module and signal collection module,software system including system control and collection module and imaging reconstruction module,and describle the entire imaging process in detail,including sample preparation,systems calibrtaion,data acquisition,data processing andreconstruction algorithms.Lastly,we have imaged the biological sample with systems and showed its imaging capabilies,we also compared the imaing results with Confocal Microscopy and Selective Plane Illumination Microscopy,and analysed some factors such as the number of projection angle,the accuracy of rotate and CCD pixel off from center which influencing the quality of reconstructed image.
