| The non-steroid anti-inflammatory drugs have been widely used in clinical medicine. But it may have some side effects by misused. As a kind of easy and simple to handle and high detection sensitivity chemical detection method, chemiluminescence method has been actively used in the fields of the detection of medical contents,the analysis of food, medicolegal expertise, agriculture and environmentology.There are two parts in this thesis. In the first part, I have summarized the clinical application, action mechanism and side effects of non-steroid anti-inflammatory drugs.And I have also summarized definition, theory, characteristic and development history of chemiluminescence, and some familiar liquid phase chemiluminescence reaction system in the first part. At last, there are the application situation and combination with other analytical equipment of chemiluminescence. The second part is the research report. I have set up some new methods to detect the content of aceclofenac,mefenamic acid, ketoprofen and diclofenac sodium, by respectively using the KMn O4-formaldehyde and KMn O4-Luminol system to, on the basis of the optimal experiment condition, which combined with the flow injection technology. And then,I have applied these new methods to analysis drug content in real tablets and capsules.The specific circumstance are as follows.1. The detection of aceclofenac: It was found that formaldehyde can react with potassium permanganate in the acidic medium and produce weak chemiluminescence,which can be sharply enhanced by the addition of aceclofenac. The experiments indicated that under optimum conditions, the chemiluminescence intensity is linearlyV related to the concentration of aceclofenac in the range 8.0× 10-2~5.0mg/L with a detection limit(3σ)of 3.0× 10-2mg/L. The relative deviation is 1.72% for 0.50mg/L aceclofenac solution in 11 repeated measurements. This method is successfully applied to the determination of aceclofenac in the tablets and capsules in the range98.1%~104.6%.2. The detection of mefenamic acid: It was found that formaldehyde can react with potassium permanganate in the acidic medium and produce weak chemiluminescence, which can be enhanced by the addition of mefenamic acid greatly. The experiments indicated that under optimum conditions, the chemiluminescence intensity is linearly related to the concentration of aceclofenac in the range 4.0×10-4~10.0mg/L with a detection limit(3σ)of 3.0×10-4mg/L. The relative deviation is 1.20% for 10.0mg/L aceclofenac solution in 11 repeated measurements. This method is successfully applied to the determination of aceclofenac in the tablets and capsules in the range 98.7%~103.3%.3. The detection of ketoprofen: It was found that Luminol can react with potassium permanganate in the alkaline medium and produce weak chemiluminescence, which can be greatly enhanced by the addition of ketoprofen. The experiments indicated that under optimum conditions, the chemiluminescence intensity is linearly related to the concentration of aceclofenac in the range 4.0× 10-2~7.0mg/L with a detection limit(3σ)of 9.0× 10-3mg/L. The relative deviation is 1.90% for 0.30mg/L aceclofenac solution in 11 repeated measurements. This method is successfully applied to the determination of aceclofenac in the tablets and capsules in the range98.1%~100.4%.4. The detection of diclofenac sodium: It was found that formaldehyde can react with potassium permanganate in the acidic medium and produce weak chemiluminescence, which can be enhanced by the addition of diclofenac sodium sharply. The experiments indicated that under optimum conditions, the chemiluminescence intensity is linearly related to the concentration of aceclofenac in the range 7.0 ×10-2~10.0mg/L with a detection limit(3σ)of 1.2×10-2mg/L. The relative deviation is1.41% for 3.0mg/L aceclofenac solution in 11 repeated measurements. This method is successfully applied to the determination of aceclofenac in the tablets and capsules in the range 97.6%~101.7%%. |
