Dechlorane Plus (DP) is a high production volume chlorinated flame retardant. It can be bioaccumulated in organisms, and has been increasingly detected in environmental matrices and biota. Toxicity data for DP are very limited till now. Our previous study demonstrated DP could increase cell proliferation in human breast cancer cell MCF 7 and induce expression of estrogen receptor and MAPK/ERK pathway related-genes. In this study, proteomic profiles of MCF 7 exposed to DP were analyzed to discover the possible molecular mechanisms of DP. Results obtained in this study will undoubtedly provide a scientific basis for toxicity and biological risk assessment of DP.The proteomic profiles were different between MCF-7 cells exposed to 10-6M DP for 96 hours and control groups. Totally twenty-three proteins were found differentially expressed with significant difference (p<0.05). Nine of them were down-regulated, while fourteen of them were up-regulated. Ten spots were cut from 2-DE gels and analyzed by MALDI-TOF MS/MS. Bioinformation analysis identified these proteins were 60kDa and 27kDa heat shock protein, two isoforms of Myosin light chain alkali, Nucleoside diphosphate kinase B,60S acidic ribosomal protein P2, Peptidyl-prolyl cis-trans isomerase A, Profilin 1, Rho guanine nucleotide exchange factor 5 and Protocadherin alpha 9 precursor. Gene oncology analysis demonstrated these proteins were mainly related to cellular process, biological regulation, response to stimulus, metabolic and development process, as well as signaling and immune system process, etc.. HSP60 was selected for further identification. The mRNA and protein expression level of HSP60 were correlated with the result of proteomic analysis. The differentially expressed proteins obtained in this study will undoubtedly be helpful for revealing the mechanism of DP.
