Purpose: Stigma maydis is gramineae zea maize plants(Zeamays L.) dry style and stigma, widely distributed in He Nan, Shan Dong, He Bei, Hei Longjiang, Jilin and other places, with depressurization, cholagogue, decreasing blood sugar level and other medicinal value. This issue mainly to study the preparation technique and quality standard of preparing stigma formula granules on the basis of its quality analysis.Methods: The first part, analyze and research on its quality using stigma as the research object. First of all, establish the content determination methods for the effective components of polysaccharide, flavonoids, celery element content in stigma. Meanwhile, in order to evaluate the quality of stigma quickly, easily and accuratly, established the quantitative analysis model of polysaccharides and flavones in stigma respectively using near infrared spectroscopy. Second, study the application of NIRS and HPLC fingerprint technology in the stigma qualitative analysis. Determine the analysis method for stigma HPLC fingerprint through investigating samples’ s different extraction methods, different extraction solvent, different chromatography conditions etc, and analyze stigma of different producing area. Qualitative identificate stigma of two origin with NIRS spectral discriminant analysis method, and compared with HPLC finger-print qualitative results. Finally, examine the content of moisture, total ash, acid insoluble ash content, water soluble extract, alcohol soluble extract, trace elements in stigma, and make provision for the content limitation of the above indices according to the determination results, preliminarily working out the quality standard of stigma. The second part, confirme the preparation method of stigma formula granules by optimizing extraction process, concentration process, drying process and granulation process. Analyze the correlative parameters of stigma formula granules refer to the pharmacopoeia rules, preliminarily working out its quality standard.Results: The first part, firstly, determine the normal analysis method of yellow ketone, polysaccharide, celery element content in stigma, simultaneously, establish an exact, rapid method to determine the content of yellow ketone and polysaccharide in stigma with near infrared spectroscopy. Secondly, establish the stigma HPLC finger-print analysis method and the near infrared qualitative analysis model, the analysis models established are able to differentiate stigma samples from two origin accurately, the classification identification accuracy of calibration set sample and the prediction identification accuracy of validation set samples are100%. Therefore, NIRS spectrum discriminant analysis results consistent with HPLC finger-print analysis results. Finally, through the study of stigma quality standards, tentatively fix that the stigma moisture should not exceed 11.38%, water soluble extract content shall not be less than 7.97%, alcohol soluble extract content shall not be less than 5.66%, the total ash content must not exceed 5.46%, acid insoluble ash content must not exceed 2.13%, major ingredients flavones, polysaccharide and celery element content shall not be less than 0.5%, 4.6% and 0.0126%. The second part, confirm the best preparation technology for stigma formula particles: cleaning and removing impurity of stigma, soak with 25 times more water for 30 min, place on the induction cooker, boil then decocted for 30 min with slow fire, the filtrate sieve through 100 mesh sieve, repeate three times, merge the filtrate, place quietly overnight, take the supernatant. Decompressing condensed into extractum with a relative density of 1.06(80 ℃), decompressing dry extractum at 0.95 MPa, 80 ℃, smash to get extractum powder. Mix extractum powder with medicinal dextrin at mass ratio 3:1, add 80% ethanol to make softwood, sieve through 20 mesh sieve, dry for 1 h at 65 ℃, the particles we get are divided and take with double screen named 1 screen and 5 screen, obtain stigma formula particles, and preliminarily working out its quality standard.Conclusion: This issue establishes the quality analysis method of the stigma, confirms the preparation technology and quality standard control of stigma formula particles, which provide a scientific basis for Stigma medicinal materials’ quality control and stigma formula particles’ industrial production.
