Because plant polysaccharide has manybiological activities, it has become a hot research topic in recent years. The extraction and purification is the basis of research on polysaccharides. In the plant polysaccharides, sesame leaves polysaccharide has good antioxidant activity. The deep study of it will bring more development of related functional products. In this paper, the sesame leaves polysaccharide extraction and purification methods are analyzed and studied, and its composition and content are analyzed, which provides a basis for research in sesame leaves polysaccharide.The first part discusses the microwave extraction technology of sesame leaves polysaccharide, using four factors and three levels orthogonal test design. The test analysis four factors that effect the extraction rate of sesame leaves polysaccharide: the microwave extraction of the material liquid ratio, microwave power, extraction time, extraction temperature. Test results show that when the extraction time is 60 minutes, extraction temperature is 75 degrees, solid-liquid ratio is 1:10, microwave power is 750 W, the sesame leaves polysaccharide has the condition rate of 4.15%.The sesame leaves polysaccharide were preliminary purified. Decolorize polysaccharide by active carbon,and through the orthogonal experiment determine the optimum process conditions of sesame leaves polysaccharide decolorizing: activate carbon dosage is 3%, decolorization temperature is 55 degrees, reaction time is 60 minutes. Under these conditions, the polysaccharide solution decolorization rate up to 83.44%. Polysaccharide retention rate is 73.21%. Protein processing of sesame leaves polysaccharide is adopted sevage method. Sesame leaves polysaccharide on protein before and after taking off the protein content were 3.31 g/mL and 1.02 g/mL, protein removal rate was 69.2%. By phenol- sulfuric acid method, before taking off the protein t polysaccharide concentration is 3.025 mg/mL, after taking off the protein polysaccharide concentration is 2.843 mg/mL, polysaccharide retention rate is 93.98%. It shows that polysaccharide losses of sesame leaves by sevage method is small.Apurify sesame leaves polysaccharide by gas chromatography, in which at least there are five kinds of ingredients: glucose, mannose, galactose and glucuronic acid, xylose. The most abundant in the determination of components is glucose, the least is the mannose.
