Total polysaccharide from Mulberry leaves (TPM) can decrease blood sugar andcure Alloxan-induced Diabetic Mice according to modern pharmacology research.Therefore, it has great value to extract, separate, purify TPM and study its correlativecharacters to develop a new kind drug in curing diabetes.Detailed studies of extracting, separating and purifying TPM are carried out inthis paper. Compared by the effect of 3 methods in extracting TPM, it is found thatusing water as solvent is better than diluted acid or alkali. Then, optimal parameters ofextracting TPM by water and separating it by ethanol precipitation are respectivelygot through orthogonal experiments. After studying macroporous resin's absorptionand desorption for polysaccharide and pigment of mulberry leaves' water extract, thepigment removal condition is established. Furthermore, one sort of homogeneouspolysaccharide from mulberry leaves is got by using G-50 and G-200 Sephadex gelfiltration one after the other. At the same time, some new methods of extracting TPMsuch as using enzyme and ultrasonic are also tried in this paper. In addition, wediscuss the effect of stepwise addition of ethanol and ethanol precipitating times topolysaccharide content. Effect of removing protein by the method of Savage topolysaccharide and protein content is also discussed in this paper. Moreover, themineral content in water extract of mulberry leaves is measured and then removed bydialysis.At last, the solubility and purity character of purified polysaccharide frommulberry leaves are examined. It is shown that the component sugars ofpolysaccharide from mulberry leaves contain glucose, galactose and arabinose by themethod of TLC and the number averages molecular weight (Mn) is 3.1×104 and theweight averaged molecular mass (Mw) is 4.3×104 by using GPC.All the research results in this paper have great theoretical and appliedsignificance to develop polysaccharide from mulberry leaves as a new kind drug tocure diabetes.
