Study And Extraction Of Phospholipid From Sesame Oil Sediment

Abstract/Summary:
During the processing of sesame oil, a lot of oil sediment was deposited at the bottom ofthe oil tank. The sediment contained various bioactive substance such as phospholipids,sesamin, sesamol et al. The objective of this research was to extract and purify sesamephospholipids from sesame oil sediment. The constituent of sesame phospholipids, thecomposition of fatty acids, the content of lignin were all detected. Refine the recovery sesameoil to abtain ediable sesame oil.Analyzed oil sediment which was made from different process, the results showed thathydraulic pressed oil sediment was the suitable material to extract phospholipids for it containedmuch phospholipids and few of insoluble impurity. The major component of hydraulic pressed oilsediment was those as follows, sesame oil92.86%, phospholipids2.79%, insoluble impurity6.99%.With the hydraulic pressed oil sediment as the material, comparing the two methods hydration,combination of centrifugal and extraction on the effects of phospholipids extracting. In theconcentrated sediment made under the optimum conditions by hydration, the content ofphospholipids was6.98%, insoluble impurity15.45%, oil84.5%. In the oil made under theoptimum conditions by method of combination of centrifugal and extraction, the content of oilwas94.79%, phospholipids3.41%, insoluble impurity0.04%.Mixed the oil from centrifugal and from extraction. With such oil as the material, theoptimum conditions of extracting phospholipids are as fellows, temperature of hydratedconcentration80℃, water addition was4times as that of phospholipids, reaction time30min.The yield was1.72%, the content of the acetone insolubility was45.13%。Prepared the powdered phospholipids by mixed solvent. Remove the impurity from theconcentrate phospholipids with hexane, add acetone into the filtrate. Added hexane at level of11mL/g to remove impurity, standing for5h, added acetone with ratio of10:1, extract3times.The yield of power phospholipids was36.32%,the content of acetone insolubility was95.47%。The major component of phospholipids was found to be PE, PA, PI, PC, the content ofthem in concentrate phospholipids(AI45.13%) was respectively10.74%,7.49%,1.71%,23.88%. In powered phospholipids(AI95.74%) was respectively12.75%,16.91%,2.98%,37.21%。The content of saturated fatty acid was29.15%, unsaturated fatty acid was69.04%.The content of sesamin in concentrate phospholipids was detected to be204.37mg/100g, sesamolin105.78mg/100g,sesamol2.09mg/100g. The content of sesamin in concentratephospholipids was59.04mg/100g, sesamolin1.19mg/100g,sesamol0.57mg/100g. Sesamin,sesamol, sesamolin were of various bioactive, these ligian would benefit the sesamephospholipids have new and better application value.Treat the recovery sesame oil with alkali refining and adsorption bleaching to abtainediable sesame oil. After alkali refining, the acid value of sesame oil was reduced to0.72, thecolour value was yellow10.4, red2.0(133.4mm cell), close to the high-grade oil; afteradsorption bleaching, the acid value of sesame oil was reduced to3.12, the colour value wasyellow19.9, red2.9(25.4mm cell), close to the second order sesame oil.
