Background: World Health Organization estimated that the number of diabetes patients aroundthe world was increasing and they were younger than before. Recently, the majority is the T2DMpatient, on which NPY in pancreas has a certain effect. The research on diabetes is focused onNPY in the tissues of the hypothalamus and plasma, but there were few studies on the effects ofacute and chronic exercise on NPY and no reports about Y1receptor protein expression inpancreas of T2DM elder rats. Objective: To research the effects of acute and chronic exercise onT2DM old aged rat pancreas, NPY and Y1receptor protein expression. It will reveal the exerciseintervention in improving T2DM pancreatic NPY resistance mechanism and provide a theoreticalbasis for the clinical development of T2DM exercise intervention schedule. Methods:12ratswere divided randomly and fed with basal diet.They were taken as the normal control groupfrom the5211-month-old male SD rats.The other40rats were fed with high fat diet,which weretaken as High-fat diet group. The model of obesity was prepared after High-fat diet for8weeks.Then the rats were divided into three groups after STZ (35mg/kg body weight) induction. Theywere diabetic control group (n=12), acute exercise group (n=14), and chronic exercise group(n=13). Results:①Serum insulin, glucose and triglycerides were decreased significantly (p <0.01)and HDL,NPY-Y1receptor protein expression were increased significantly (p <0.01) in the acuteexercise and chronic exercise groups compared with the normally controlled group.②Seruminsulin, glucose, triglycerides, LDL, and NPY protein expression in pancreas were decreasedsignificantly (p <0.01), HDL and NPY-Y1receptor protein expression were increased clearly(p <0.05) in the acute exercise and chronic exercise groups compared with the diabeticgroup.Conclusions:①T2DM can produce insulin resistance and glucose and lipid metabolismdisorder,which may be associated with pancreatic NPY resistance.②Acute exercise and chronicexercise can reduce NPY protein expression in pancreas and improve its sensitivity,up-regulation NPY-Y1receptor protein expression and NPY sensibility NPY resist and theeffects of acute exercise can last for about16hours. Advices: It needs research further aboutacute exercise after16hours of T2DM rat pancreatic NPY and its receptors influence.
