Objective:To study the effect of nAchRα9Antibody on the acantholysis of pemphigusvulgaris through establishing mouse model, In order to provide a reference basisto further clarify the mechanism of pemphigus.Methods:The Balb/c neonatal mice were divided into16groups,a total of160.Miceof group1-5were given subcutaneous injection of the serum of PV patients toestablish mouse model; the newborn mice were killed12h,18h,24h,36h and48h after injection respectively. Mice of group6-10were given subcutaneousinjection of the nAchRα9Antibody; the newborn mice were killed12h,18h,24h,36h and48h after injection respectively. Mice of group11-15were givensubcutaneous injection of the serum of PV patients and nAchRα9antibodysimultaneously; the newborn mice were killed12h,18h,24h,36h and48hafter injection Mice of group16were given subcutaneous injection of normalhuman serum; the newborn mice were killed48h after injection respectively.respectively. The effect of nAchRα9antibody on PV acantholysiswas evaluated in terms of clinical manifestation, histopathology and directimmunofluorescence.Results:acantholysis was not observed in the neonatal Balb/c mice of group6-10injected with nAchRα9antibody alone;the neonatal Balb/c mice of group1-5andgroup11-15injected with the serum of PV patients,As for the neonatal mice ofgroup1and group11were killed12h after injection, acantholysis was notobserved in group1, only1case of acantholysis in group11was observed;as forthe neonatal mice of group2and group12were killed18h after injection, group12showed higher degree of acantholysis than group2;as for the neonatal miceof group3and group13were killed24h after injection, group13showed higherdegree of acantholysis than group3;as for the neonatal mice of group4andgroup14were killed36h after injection, the acantholysis degree showed nosignificant difference between group4and group14(P>0.05);as for the neonatalmice of group5and group15were killed48h after injection, the acantholysisdegree showed no significant difference between group5and group15(P>0.05);acantholysis was not observed in the neonatal Balb/c mice of control groups ofgroup16.Conclusion:1.Using the serum of PV patients and nAchRα9antibody to inject Balb/cneonatal mice simultaneously can promote the generation of acantholysis andincrease the degree of acantholysis in PV, which prompt that nAChRα9antibodymay play a role in promoting the onset of PV;2.nAchRα9antibody alone hardly induces acantholysis,it must with the other autoantibody of PV-IgG and then can induce PV change. Objective:To study the effect of anticholinesterase agents on the acantholysis ofpemphigus vulgaris through establishing mouse model, offer the animalexperiment basis to the feasibility of anticholinesterase agents to treat withpemphigus of glucocorticoid-free.MethodsBalb/c neonatal mice were divided into4groups, a total of55.The neonatalBalb/c mice of group1were injected subcutaneously with the serum of PVpatients to establish PV mouse model.On the basis of serum injection,group2and group3were injected with Pyridostigmine Bromide and NeostigmineMethylsulfate respectively.The neonatal Balb/c mice of group4were injectedsubcutaneously with normal human serum as control. The effect ofanticholinesterase agents on the acantholysis of PV was evaluated in terms ofclinical manifestation, histopathology and direct immunofluorescence. ResultsThe injection of PV patients’ serum alone induced PV changes in Balb/cneonatal mice of group1.The degree of acantholysis induced by PV patients’serum was reduced in group2and group3. No changes were observed in thecontrol Balb/c neonatal mice of group4.ConclusionPV patients’ serum can succeed in inducing the changes of PV in Balb/cneonatal mice,inject with Anticholinesterase agents simultaneously,no matterPyridostigmine Bromide or Neostigmine Methylsulfate,anticholinesterase agentscan reduce acantholysis of PV,and it might have a therapeutic effect onpemphigus vulgaris...
