Objective:To observe the effect of uric acid at different concentrations on the neural differentiation of bone marrow mesenchymal stem cells.Methods:In vitro, hBMSCs were isolated, purified and cultured. The morphology change was observed. The third passages of hBMSCs were cultured in the standard serum-containing medium and induced to differentiate to neuron-like cells by induced liquid containing four different concentrations of uric acid (0mmol/L as control group,0.2mmol/L,0.4mmol/L,0.8mmol/L). After intervention, cells were detected by Nissl staining and specific markers were detected by immunohistochemistry method.Results:hBMSCs were obtained successfully by adherent method. Cells in the standard serum-containing medium group were long spindle. In the control group and the induced group, after induction, cell body shrank, forming processes and connections. Nissl body was found. The positive rates of NSE and NF-M significantly were higher compared to the control group (P<0.05). The positive rates of NSE and NF-M were increased as the increase in concentration of uric acid (P<0.05). The positive rates of Nestin were decreased compared to the control group (P<0.05), moreover, the positive rates of Nestin were decreased as the decrease in concentrations of uric acid (P<0.05). After4hours of induction, cells fell off significantly.Conclusion:In a certain period of time, uric acid can promote BMSCs into neuron-like cells, and has a certain concentration-dependent manner in vitro.
