Cardiovascular and cerebrovascular diseases is a kind of common disease that seriously threatens human beings,especially middle-aged and senior people over50-years-old. Every year the number of people died of cardiovascular and cerebrovascular diseases reaches up to about15million, ranking first among all causes of death. Cardiovascular and cerebrovascular diseases has already become the top killer in the list of causes of death, characteristic of high morbidity, high disability rate, high mortality, high recurrence rate and many complications. Therefore, it is one of the most significant public health problems that we need face and solve for a long time both presently and in the future. As atherosclerotic plaque gets into the unstable state from the stable state, plaque ruptures, and enhanced platelet aggregation and adhesion bring about thrombogenesis and angiemphraxis, regarded as a common pathological basis on acute cardiovascular and cerebrovascular diseases. As a result, it is of great importance and interest in atherosclerosis (AS) prevention research in the field scientific research all along.AS is a complex multifactorial pathological process, and therefore a single mechanism of action of the drug is probably not fully inhibit the development of AS. The Chinese medicine is based on traditional Chinese medicine theory, focusing on the macro level, comprehensive treatment, overall adjustment, in theory, from the multi-angle, multi-level, multi-target effective prevention and treatment of AS. In recent years, some scholars believe that atherosclerosis within the scope of Chinese medicine "Pulse turbidity" disease, blood turbidity blocking collaterals is its pathogenesis characteristics. TCM Resolving turbidity and Dredging collaterals method is established for this syndrome, which has received a good clinical efficacy. In this study, starting from the perspective of prevention, aimed proven Huazhuotongmai anti AS mechanism from the aspects both in inflammation and blood lipid through animal experiments.Objective:Through animal experiments to explore the effects of Huazhuo tongmai compound on blood lipids, inflammatory factors and atherosclerotic plaque of high-fat diet-induced AS rabbit model.Method:40male New Zealand Rabbits are divided into4groups at random:blank control group, model group, simvastatin group and Huazhuotongmai group. Except blank control group, every rabbit was fed by high fat diet for15weeks in order to copy rabbit model of AS. Administrating while modeling conducted for15consecutive weeks. Before dosing and administration for14weeks after, blood smaples were drawn via ear marginal vein to determine the total cholesterol(TC), triglyceride(TG),high-density lipoprotein(HDL), low-density lipoprotein(LDL), Apolipoprotein AI(ApoAI), apolipoproteinB(ApoB), C-reactiveprotein(CRP), Interleukin-2(IL-2), Interleukin-6(IL-6),tumor necrosis factor-a (TNF-a). After15weeks, observed the morphological changes of Thoracic aorta using HE and oil red O staining.Results:(1)Lipids testing:14weeks after administration, compared with the blank control group, model group TC, TG, HDL, LDL,ApoAI, ApoB were significantly increased (P<0.01). Compared with the model group, the statin group TC, TG, LDL, ApoB decreased with statistical significance (P=0.01,P=0.006,P=0.000, P=0.004, P<0.05). Compared with the model group, compound group TC, TG, LDL, ApoB decreased with statistical significance (P=0.006,P=0.037,P=0.000,P=0.002, P<0.05), and effectiveness had no significant difference compared with simvastatin(P>0.05).(2)Inflammatory Factors testing:Compared with the model group, the statin group CRP, IL-2were significantly lower (P=0.023,P=0.002,P<0.05). Compared with the model group, compound group CRP, IL-2were significantly lower (P=0.022,P=0.020, P<0.05), and effectiveness had no significant difference compared with simvastatin (P>0.05).(3) Rabbit thoracic aorta HE staining(×100):In blank control group, the rabbit arterial endothelial cells intact, intimal smooth, no subendothelial lipid deposition, clear layers of the structure. In model rabbits artery intima thickening, partial deletion of endothelial cells, smooth muscle cell disorders, there are a lot of foam cell accumulation. In simvastatin group showed no rabbit arterial intimal thickening, a small amount of smooth muscle cell proliferation, arrange the basic rules, clear layers of the structure, pathological changes were significantly lighter than the model. In compound group, rabbits artery intima less smooth, a small amount of uplift, endothelial cell integrity, clear layers of the structure, pathological changes compared with the model group was significantly lighter. This suggested that AS model were made successfully and Huazhuo tongmai compound had certain intervention effects on atherosclerosis in rabbits. (4) Rabbit thoracic aorta Oil Red O staining:The blank control group showed no rabbit arterial plaque, no subendothelial lipid droplets; model group showed a large number of subendothelial lipid droplets and obvious plaques; the statin group and compound group showed a very small amount of subendothelial lipid droplets and no significant plaque.Conclusion:(1) Huazhuo tongmai compound is capable of inhibiting the formation of plaque in rabbit aorta, and reducing the subendothelial lipid deposition. Compared with simvastatin, its effectiveness has no significant difference.(2) Huazhuo tongmai compound has significantly reduced TC, TG, LDL, ApoB, and not inferior to simvastatin. Suggesting that regulate lipid metabolism may be one of the anti-AS pathways.(3) Huazhuo tongmai compound has significantly reduced the CRP and IL-2, and not inferior to simvastatin. so we can deduce its anti-inflammatory effect may be one of the anti-AS pathways.
