The World Health Organization predicts that by the year2030, the global diabetes will reach the number by366million, of which the proportion of type2diabetes is up to95%. Currently,the treatment of type2diabetes is mainly based on western medicine, but many complications and side effects in patients after the western medicine treatment of type2diabetes are the major problems that the medical workers and researchers are facing with, this is a huge challenge.But meanwhile,it is also a good opportunity for the development of Chinese medicine,for it’s great role of providing a broader space for the development of diabetes’ Chinese traditional treatment.Acupuncture prevention of diabetes and its complications in China has a history of more than two thousand years. In recent years, acupuncture seems to be more widely used in clinical treatment and care of diabetes, and it had achieved good results. Yishu point is an extra acupoint, which is closely related with the pancreas. Some scholars believe that Yi shu point is the location of pancreas qi gathers, so it has the effect of promoting pancreatic function and improving insulin secretion, regulating blood glucose.Literature data showed that in the clinical acupuncture treatment of diabetes, Yishu point is mainly used as the chief point and accompanied with other acupoints, although the literature showed acupuncture had obvious effect of lowering blood glucose, but there was rarely study for the therapeutic effect of the single Yishu point in both clinical and animal experiments study. Therefore, in our study,we adopted "Yishu" point to treat type2diabetic rats model, to study the effect of "Yishu" for lowering blood glucose of the type2diabetic rats model after the interventionObjective:Observe the regulation of electro-acupuncture (EA) of "Yishu" point of type2diabetic rats’ pro-insulin mRNA expression in pancreas and EA of "Yishu" point’s impact on blood glucose, leptin,resistin and adiponectin; verify the treatment effect of type2diabetes by EAof "Yishu" point; state the key role of electro-acupuncture during the treatment; reveal the possible mechanism of the EA of "Yishu" point for type2diabetes.Method(1) Acorrding to the random number table,we randomly put8rats into nomal control group,and these rats were raised with nomal fodder,the left50ones were used for modeling. The method of high-fat-diet manipulation combined with low-dose streptozotocin (STZ) intraperitoneal injection was adopted to establish type2diabetic rat model, after the modeling,the random number table was used again to divide the successful T2DM rats model into model control group, treatment group of EA"Yishu" point, treatment group of EA "Pishu" point, treatment group of EA"Shenshu" point;(2) Real-time PCR method was used to detect pancreatic tissue mRNA expression of pro-insulin level;(3) Type2diabetic rats’ fasting blood glucose was measured by Roche glucometer;(4) Radioimmunoassay was employed to check insulin, leptin, resistin and adiponectin content in serum;(5) HE staining method was used to observe the pathological changes in pancreatic tissue, the expression average optical density(AOD) of positive insulin expression in pancreatic cells was detected by immunohistochemistry.Results(1) Using the method of high-fat-diet manipulation combined with low-dose streptozotocin (STZ) intraperitoneal injection can successfully duplicate type2diabetic rats model, we took50rats for modeling,42rats were successfully induced, and the success rate of modeling was84%.(2) Compared with normal control group, the pro-insulin mRNA expression of model control group was significantly lower (P<0.01); Compared with the model control group, the pro-insulin mRNA expression in the three treatment groups were increased significantly (all P<0.01), the treatment group of EA"Yishu" point increased particularly evidently;(3) Compared with the normal control group, rats’ blood glucose in the model control group was significantly increased(P<0.01),and body weight was significantly decreased after the modeling(P<0.01); After the electro-acupuncture treatment,the blood glucose in rats of treatment group of EA"Yishu" point and EA"Pishu" point declined, treatment group of EA"Yishu" point decreased more significantly (P<0.01);(4) Compared with normal control group, rats’ serum insulin level of the model control group increased, but the difference had no statistical significance (P>0.05), the three treatment groups also decreased,and the treatment group of EA"Tishu" point and EA"Shenshu" point were with very significant difference (all P<0.01); Compared with normal control group, rats’ serum leptin, resistin, adiponectin content in the model control group were lower, there were significant differences (all P<0.05);(5) Compared with the normal control group,rats’ insulin average optical density (AOD) in pancreas of the model control group was significantly lower (P<0.05); Compared with the model control group, the treatment group of EA"Yishu" point and EA"Pishu" point were significantly increased, especially for the treatment group of EA"Yishu" point (respectively, P<0.01,P<0.05).Conclusion (1) The modeling method we adopted was practicable,by which the established type2diabetic rat model was stable;(2) The possible mechanism of reducing rats’ blood glucose by EA of "Yishu" point:"Yishu" point and the pancreas are both dominated by sympathetic T6-T10spinal segments,"Yishu" point is in line with the principle of nerve segment domination. In the nerve segment domination of "Yishu", the spinal cord is considered as a primary hub, not only receives signals from acupuncture of "Yishu " point, but also receives pathological signals from the pancreas, spinal cord provides a forum for the integration of these two signals. In addition,EA of "Yishu" Point can also possiblly act through the neuro-endocrine-immune regulation pathways. EA of "Yishu" point can improve the dysfunction of type2diabetic rats’ pancreatic β cell for some extent, enhance expression of the pro-insulin mRNA, increased insulin sensitivity, reduce insulin resistance, decrease blood glucose.In summary, EA of type2diabetic rats’"Yishu" point of the type2diabetic rats can increase pancreatic insulin mRNA expression at the gene level, making insulin mRNA expression developped positively; For the cellular and molecular level, it can improve the function of islet β cells and promote the recovery of damaged cells, which helps to improve the normal function of insulin for controlling blood sugar and decrease insulin resistance which is also one of the important mechanisms for the EA of "Yishu" point to lower the blood sugar.
