Objective:To investigate the effect and mechanism of Danshen injection combined with doxorubicin in the treatment of breast cancer.Method:By erecting the nude mouse transplanted tumors models from breast cancer, randomly divided the40nude mice into Danshen injection combined with doxorubicin、doxorubicin、Danshen injection and model groups, according to the dosage regimen to medicine, and recorded the nude mouse living condions and body weight changes, dynamically observed the transplanted tumors growing conditions、measured the tumors volume and drew the tumors growth curve. Executed the mouse after10days medicine, weighted the tumors quality and calculated the tumors inhibition rates. Meanwhile, Checked blood routine, liver and kidney function. Furthermore, detected the MVD and VEGF of the transplanted tumors by immunohistochemical method.Results:1. On the average weight of nude mice, the Danshen injection combined with doxorubicin group and the doxorubicin group had significantly lower than that of the Danshen injection and model groups (P<0.01); meanwhile, the average body weight of the Danshen injection combined with doxorubicin group was slightly higher than the doxorubicin groups (P<0.05); 2.On the transplanted tumors weight, the the Danshen injection combined with doxorubicin group and the doxorubicin group had significantly lower results compared with the model group (P<0.01); the Danshen injection combined with doxorubicin group had lower results compared with the doxorubicin group (P<0.05).3.On the number of white blood cells, the Danshen injection combined with doxorubicin group and the doxorubicin group were significantly lower than the model group (P<0.01); the Danshen injection combined with doxorubicin group was slightly higher the doxorubicin group (P<0.05). Compared with the model group, the platelet count of Danshen injection group and the Danshen injection combined with doxorubicin group decreased significantly (P<0.01); meanwhile, there was no significant difference among the doxorubicin group and the model group in platelet count(P>0.05).4.On the influence of the ALT, AST, Cr and BUN,the Danshen injection combined with doxorubicin group and the doxorubicin group were significantly higher than those of the model group (P<0.01); moreover, the Danshen injection combined with doxorubicin group was less than the doxorubicin group (P<0.05).5. The microvessel density(MVD) in the Danshen injection combined with doxorubicin group and the doxorubicin group was much lower than that of the model group(P<0.01); the Danshen injection combined with doxorubicin group had lower MVD value compared with the doxorubicin group(P<0.05).6.On the VEGF expression of the transplanted tumors, the Danshen injection group can lower expression of VEGF compared with the model group(P<0.05); the Danshen injection combined with doxorubicin group and the doxorubicin group had significantly lower average value compared with the model group (P<0.01); the Danshen injection combined with doxorubicin group had lower average value compared with the model group (P<0.05).Conclusion:1. The Danshen injection combined with doxorubicin can improve the efficacy of chemotherapy.2. The Danshen injection combined with doxorubicin can reduce the damage effect of the doxorubicin on white blood cells, liver and kidney function.3. The Danshen injection combined with doxorubicin treatment can effectively down-regulate the expression of VEGF and MVD,then inhibit the angiopoiesis, which is one of the tumor-inhibiting mechanism of improving the curative effect of doxorubicin in the treatment of breast cancer.
