Objective:preparation of intestinal flora biotransformation model was used by the fresh stool of healthy volunteers. The model has been used to transform the Pulsatilla chinensis water extract of traditional Chinese medicine. By comparison prior to conversion, after Pulsatilla chinensis water extract of the main chemical components and pharmacological effects, analysis of the intestinal flora biological transformation of traditional Chinese medicine Pulsatilla chinensis composition and activity. By comparison prior to conversion and post-conversion Pulsatilla water extract of the main chemical components and pharmacological effects of changes to the analysis of the intestinal flora biotransformation of Chinese medicine Pulsatilla composition and activityMethods:(1) The fresh excreta of healthy volunteers was prepared for the intestinal microflora biotransformation model, the model was incubated with Pulsatilla chinensis water extracts under anaerobic conditions, and the transformed Pulsatilla chinensis extract was produced.(2)Pulsatilla saponin B4changes in Pulsatilla chinensis extracts between prior to conversion and after the transformation were determined by high performance liquid chromatography (HPLC).(3) The changes inhibitory effect in vitro between two kinds of Pulsatilla chinensis extraction was observed by agar diffusion method and dilution method, and MTT assay was used to compared the inhibition between human hepatoma cells (Hep-G2) and human gastric cancer cells (SGC-7901). Macrophage phagocytic function and mouse intestinal fecal SIgA levels was examined for the changes of intervention effects on gastrointestinal mucosal immunityResults:(1) High performance liquid chromatography test results show that there are significant differences in the two extract content of Pulsatilla saponin B4.(2)Vitro antibacterial experiments, The Pulsatilla chinensis water extracts can inhibit the growth of Shigella, Staphylococcus aureus, Escherichia coli, Bacillus subtilis, Pseudomonas aeruginosa. In antitumor experiments, compared with the blank control group, the two experimental groups for both tumor cells was inhibited, the difference was statistically significant (P<0.05or P<0.01). The inhibition rate has been enhanced by the transformation of the intestinal microflora of Pulsatilla chinensis water extract for the two tumor cell than before the transformation, the difference was statistically significant (P<0.05). In macrophage phagocytosis neutral red experiments, compared with the blank control group, the two experimental groups could increase the rate of macrophage phagocytosis, and the difference was statistically significant(P<0.01). The pulsatilla chinensis water extracts converted by the intestinal microflora group the macrophage phagocytosis rate higher than before the transformation, the difference was statistically significant (P<0.05). The detection results of the SIgA in mouse intestinal mucosa, that compared with the blank intestinal bacterium group, two experimental groups of mice intestinal mucosal SIgA level has increased, and the difference was statistically significant (P <0.05). The mice intestinal mucosal SIgA level of Pulsatilla chinensis water extract after the transformation has increased than before the transformation, and the difference was statistically significant (P <0.05).Conclusion:The feces incubated method can be successfully prepared the intestinal microflora transformed model. The the Pulsatilla chinensis water extracts transformed by intestinal microflora, its chemical composition and its anti-bacterial, anti-tumor, the intervention of the immune function of the intestinal mucosa in varying degrees change.
