Objective:To study the relationship between hypothalamic of sleepYang Deficiency rats in the hypothalamus and apoptosis gene (BCL-2, bax)with dajianzhong decoction and the pain. From the level of genetic study of the dajianzhong decoction in the treatment of spleen Yang deficiency and the mechanism of abdominal pain, and for the clinical application of the lunar month of dajianzhong decoction and development has laid a solid theoretical and experimental basis for the party, the research provides a new idea and method.Methods:Sixty SD rats were divided into6groups, each group with ten rats;they are normal control group, model group, high-dose group with dajianzhong decoction, middle-dose group with dajianzhong decoction, low-dose group with dajianzhong decoction and western medicine control group. In addition to the normal control group, the rest of the group are through eating disorders, Laojuan excessive, from the bitter cold to establish the rat model of spleen Yang deficiency, and after intraperitoneal injection of acetic acid solution method, establishes the spleen Yang deficiency rats pain model. After the success of the model, in addition to the normal group and model group were given the lunar month of30days in high dose group of soup, soup in the lunar month of30days small doses group corresponding decoction irrigation uniform, western medicine indomethacin control group give indomethacin aqueous solution, the treatment after15days, respectively take6group rats organization, using immunohistochemical method, observation in the hypothalamus Bcl2and Bax gene positive expression.Results:the spleen Yang deficiency rats in the hypothalamus Bcl-2express decrease, Bax express obviously increased, and compared with normal group (P<0.01), lunar month of30days of soup can obviously improve the rat spleen Yang deficiency syndrome, make the rats in the hypothalamus Bcl-2gene expression significant rise, Bax gene positive expression significantly reduced compared with model group,(P<0.01), high dose group of the lunar month of30days soup, western medicine control group, no significant difference between them (P>0.05). Lunar month of30days in small, medium dose group soup rats in the hypothalamus Bcl-2express although have rise, Bax express although have decreased, but no big dose significantly, compared with the amount of obvious effect relations (P<0.01).Conclusion:treatment with dajianzhong decoction, spleen Yang deficiency rats with abdominal pain of the disease obviously improved and its mechanism may be through the lower one of Bax positive expression, rise Bcl-2expression, inhibit apoptosis, reduce hypothalamus tissue damage, directly or indirectly, to improve the purpose of the spleen Yang deficiency syndrome.
