Objective:This study through the establishment of respiratory syncytialvirus pneumonia age in BALB/c mice model,Using in vivo animal testing,Researchyupinfengsan extract and yinqiaosan extract therapy effect on RSV pneumoniainduced the young mice.Discuss it on RSV infection in the expression level ofIL-4,TLR4. provide experimental basis for clinical prevention and treatmentof RSV.Materials and methods:1.Using nasal immunization folded respiratory virus long of RSV infection inyoung mice model was established, through breathing, food, fur, active andgeneral state changes such as body weight, the lung index determination,the lungtissue appearance,and the lung tissue pathology under light microscopyobservations to identify RSV pneumonia in mice model.2.RSV infection model was established,combined with the general state,pathological lung tissue of mice, morphology, the lung index indicators, observeyupinfengsan extract and yinqiaosan extract prevention and cure function onRSV pneumonia in mice.3.Using immunohistochemical method,detect the TLR4protein expression in lungtissue of mice in different time points by administration of the extracts ofYupingfengsan and Yinqiaosan.4.Using ELISA method,,detect the TLR4protein expression in serum of mice indifferent time points by administration of the extracts of Yupingfengsan andYinqiaosan.observe the role of prevention and treatment of the extracts ofYupingfengsan and Yinqiaosan five flavors to IV pneumonia in mice.Results:1.After RSV virus vaccination, normal group basic keep in good condition, modelgroup, fur, breathing, eating, etc. General state changes,lung tissue pathologyand appearance are changed,compared two groups of mice lung index had significant difference in observation cycle(P <0.05).2.Mice lung index according to the results at different times:The first dayand seventh day after building,compared with the normal group,model group havesignificant differences(P <0.05);compared with model group, shuangbiaogroup,jiebiao group and ribavirin group have the significant difference (P <0.05). third and fifth day after building, compared with normal group, the restof the group had significant difference (P <0.05); compared with model group,the rest of the group have the significant difference (P <0.05).3.Lung tissue TLR4protein expression results show that:building on the firstday, compared with the normal group,the model group had significant difference(P <0.05).building, third, five days after, compared with the normal group,the rest of the group have the significant difference (P <0.05);compared withmodel group, shuangbiao group,jiebiao group and ribavirin group have thesignificant difference (P <0.05); building after7days, compared with thenormal group,model groups had significant difference (P <0.05); compared withmodel group, the rest of the group had significant difference (P <0.05); comparedwith ribavirin group, model group there were significant differences (P <0.05).4.Peripheral blood content of IL-4shows:building on the first day, comparedwith the normal group,the model group had significant difference (P <0.05).building, third, five days after, compared with the normal group, the restof the group have the significant difference (P <0.05);compared with model group,shuangbiao group,jiebiao group and ribavirin group have the significantdifference (P <0.05); building after7days, compared with the normalgroup,model groups had significant difference (P <0.05); compared with modelgroup, the rest of the group had significant difference (P <0.05); comparedwith ribavirin group, model group there were significant differences (P <0.05).Conclusion:1.The extracts of Yupingfengsan and Yinqiaosan can improve the respiratorysyncytial virus infection general status in mice, decreased lung index, reduce the pulmonary pathological changes.2.Shuangbiao method can reduce inflammation of the lung injury in mice, lowerlevels of IL-4and TLR4, prevent virus damage in lung tissue.3.Yinqiaosan junchen extract and yupingfeng extract is an effective drug fortreatment of respiratory syncytial virus infection, its action mechanism maybe through adjust the level of IL-4, TLR4play a role.
