Objective:This experiment use the traditional Chinese medicine therapy-acupoint catgut embedding therapy to treat optic nerve injury model in rats and observe the changes of rat retinal ganglion cells and the relationship between the changes and damage time. In this way can provide laboratory basis for mechanism of acupuncture treatment for traumatic optic nerve injury.Methods:208rats are randomly divided into6groups:blank group, control group, model group, acupoint catgut embedding therapy group, neurotrophic factor group and acupoint catgut embedding therapy with neurotrophic factor group. Among the last five groups, they are divided into5subgroups:4days group,7days group,14days group,21days group and28days group. Making the traumatic optic neuropathy rats with transactional injury. After catgut embedding on the Ganshu (BL18), observing the morphology of retina and numbers of the retinal ganglion cells, and detecting the expressing situations of nestin and GAP-43in LGN with immune histochemical method after therapy.Results:Compared with the model group, the retinal ganglion cells are rising in acupoint catgut embedding therapy group, neurotrophic factor group and acupoint catgut embedding therapy with neurotrophic factor group (P<0.05or P<0.01). Compared with the model group, the expression of nestin and GAP-43in LGN is higher in acupoint catgut embedding therapy group, neurotrophic factor group and acupoint catgut embedding therapy with neurotrophic factor group (P<0.05or P<0.01).In the early damage, the number of RGC in acupoint catgut embedding therapy group is more than neurotrophic factor group (P<0.01). But14days later, the results are just the opposite.Conclusion:1. acupoint catgut embedding therapy is an effective treatment in treating the optic nerve injury rats. Using the acupoint catgut embedding therapy, The early effect is effective. But after14days neurotrophic factor is better.2. We speculated that the mechanism of treating TON by acupoint catgut embedding therapy is reducing the mortality of RGC, raising the expression of nestin and GAP43. Activate the neural stem cells in the central nervous system inorder to promote the growth of axons.3. When treating TON, combine traditional Chinese and western medicine treatment is not as good as point embedding therapy or neurotrophic treatment.
