Comparative study on effect of Shengmai Decoction and Shixiao powder ofAdriamycin cardiotoxicity in rats model of myocardial energy metabolismObjective Through observating similarities and differences on effect of the two kinds of Chinese classical medicine prescriptions:Shengmai Decoction and Shixiao powder of myocardial energy metabolism with Adriamycin cardiotoxicity rats model,comparative study on influence and mechanism of the two treatment prescription:Supplementing Qi and nourishing yin and Promoting blood circulation and removing blood stasis.Methods Randomly divided108male SD rats of clean grade into6groups:Normal group, Model group,Trimetazidine group,Shengmai Decoction high-dose group,Shengmai Decoction low-dose group, Shixiao powder group(n=18).Intraperitoneal injection of Adriamycin2.5mg/kg,once a week,6times totally, the cumulation of15mg/kg.Normal group were injected equal saline daily,schedule as the above mothed. after model were established,the corresponding me-dicine were given (trimetazidine group,Shengmai Decoction low-dose group,Shixiao powd er group were given clinically equivalent dose,Shengmai Decoction high-dose group were given four times of clinical dose),once daily.saline was given to Normal group.14days, half number were taken samples,28days, the remaining as the above. Serum SOD,MDA;expression of PPARa mRNA;bcl-2and Bax protein were tested.Results①the ELISA method:14days,SOD level of Model group was significantly lower than other groups(P<0.05);compared with Model group,drug groups could improve SOD (P<0.05),Trimetazidine was most significant;Shengmai Decoction high-dose group was superior to low-dose group(P<0.05);Shengmai Decoction low-dose group was higher than Shixiao powder group(P<0.05);28days,drug groups significantly differed from Model group (P<0.05);Shengmai Decoction high-dose group and Trimetazidine group showed no difference(P>0.05);Shengmai Decoction low-dose group was better than Shixiao powder (P<0.05);the SOD level28days of Model group was significantly lower than which14days (P<0.05);drug groups28days effect were better than14days(P<0.05).14days,MDA level of Model group were higher than rest groups obviously(P<0.05); compared with Model group,drug groups reduced level of MDA,Trimetazidine effect was most obvious (P<0.05);Shengmai Decoction high-dose group was better than rest traditional Chinese medicine groups(P<0.05);Shengmai Decoction low-dose group were better than Shixiao powder group(P<0.05).28days,Trimetazidine was better than Shengmai Decoction high-dose group(P<0.05);ShengmaiDecoction high-dose group was superior to low-dose group(P<0.05); Shengmai Decoction low-dose group and Shixiao powder group showed no difference (P>0.05).MDA level14days of Model group was significantly higher (P<0.05);drug groups28days were better than which14days (P<0.05).②The RT-PCR mothed:14days,the expression of Model group PPARa mRNA was lower than other groups(P<0.05);Shengmai Decoction high-dose group improved expression of PPARa mRNA,but which was weak than Trimetazidine(P<0.05);Shengmai Decoction low-dose group was better than Shixiaosan group(P<0.05).28days,drug groups significantly enhanced expression of PPARa mRNA(P<0.05);Shengmai Decoction high-dose group and Trimetazidine group showed no difference(P>0.05);Shengmai Decoction High-dose group was better than low-dose group(P<0.05); Shengmai Decoction low-dose group and Shixiao powder group showed no difference(P>0.05); the expression of Model group PPARa mRNA28days was significantly lower than which14days(P<0.05);effect of other drug groups28days were better than14days (P<0.05).③The Immunohistochemical method:14days,Bax expr ession of Model group was significantly higher than Normal group(P<0.05),Bcl-2expression was no difference(P>0.05); compared with Model group,drug groups reduced expression of Bax significantly (P<0.05), increased Bcl-2expression(P<0.05),Trimetazidine was most obvious;Shengmai Decoction high-dose group was superior to low-dose group(P<0.05); Shengmai Decoction low-dose group was better than Shixiao powder(P<0.05);Enhanced Bcl-2/Bax, Trimetazidine was most obvious;Shengmai Decoction high-dose group was better than low-dose group(P<0.05); Shengmai Decoction low-dose group was better than Shixiao powder group(P<0.05).28days,Bax expression of Model group was signifcantly increased than Normal group(P<0.05),expression of Bcl-2was no difference(P>0.05);Compared with Model group, drug groups were significantly decreased expression level of Bax(P<0.05),increased expression of Bcl-2(P<0.05);Shengmai Decoction high-dose group were better than low-dose group(P<0.05);Shengmai Decoction low-dose group was better than Shixiao powder group (P<0.05);Enhanced Bcl-2/Bax, Trimetazidine was most obvious(P<0.05);Shengmai Decoction high-dose group was better than low-dose group(P<0.05);Shengmai Decoction low-dose group was better than Shixiao powder group(P<0.05);compared with14days,Normal group28days was no difference(P>0.05);Model group expression of Bax was significantly increased(P<0.05), Bcl-2had no change(P>0.05);drug groups effect28days were better than which14days in reducing expression of Bax and increasing expression of Bcl-2(P<0.05).Conclusion1.Shengmai Decoction and Shixiao powder coule reduce oxidative stress by increasing serum SOD and reducing MDA; Shengmai Decoction was better than Shixiao powder;2.Shengmai Decoction and Shixiao powder could improve expression of myocardial PPARa-mRNA,improve myocardial energy metabolism by mediating lipid metabolism,the effect was more obvious with prolongation of drug administration;3.Shengmai Decoction and Shixiao powder could reduce protein expression of Bax,increase protein expression of Bcl-2, improving the ratio of Bcl-2/Bax to prevent apoptosis,Shengmai Decoction was better than Shixiao powder;4.Shengmai Decoction and Shixiao powder could protect Adriamycin cardiotoxicity of myocardial energy dysbolism,Shengmai Decoction high-dose was superior to Shengmai Decoction low-dose,existed dose-effect relationship,Shengmai Decoction of Supplementing Qi and nou-rishing yin was better than Shixiao powder of promoting blood circulation and removing blood stasis.efficiency of28days was superior to efficiency of which14days.
