Experimental Study On Tongsaimai Tablet On Establishment Of Collateral Circulation In Posterior Limb Ischemia Rats

Objective: To observe the effect of Tongsaimai Tablet on Establishment of Collateral Circulation in posterior limb ischemia rats.Methods: Fifty SD rats (twenty-five female rats and twenty-five male rats) were divided into Tongsaimai Tablet high dose group (12.44g crude drug/kg), Tongsaimai Tablet low dose group (6.22g crude drug/kg)、positive control group (Danshen root Tablet 0.2025g/kg), model control group, blank group at random, each group has ten rats. Established ischemia animal models in the left posterior limb femoral artery ligation except blank group. From the next day ofpostop, gave the rats drugs(10ml/kg) timed for twenty-five days. Measured the body weight of the rats and the right and left posterior limb temperature head of skin before administration, after administration the fifth day, the tenth day, the fifteenth day, the twenty day, the twenty-fifth day. And observed living status of them. After administration the twenty-fifth day, measured the platelet packing fraction, hemorheology and observed Angiogenesis in angiography before put the rats to death. After the death, took appendicular muscles of the rats to make specimens to observe blood vessel densities and pathology examination.Results: 1、weight and general status: limbs ischemia would induce body weight decrease, the difference between model control group and blank group had statistical significance. 2、ligated member status: the rats skin temperature heads of medication administration team were all lower than model control group, Tongsaimai Tablet can improve limb ischemia symptom significantly (P＜0.05). 3、rats platelet packing fraction: TongSeMai Tablet can degrade platelet packing fraction effectively. 4、hemorheology: Tongsaimai Tablet can degrade blood viscosity, and the difference had statistical significance (P＜0.05～P＜0.01). Tongsaimai Tablet high dose can degrade plasma viscosity, and the difference had statistical significance (P＜0.01). 5、arteriography: the rats of medication administration team all had left femoral artery mutilation, the rats blood vessel densities of Tongsaimai Tablet high dose group increased. 6、pathology observation: Tongsaimai Tablet high dose can improve limbs ischemia symptom obviously, muscle fiber blood vessel sum of the rats in this group were more obviously than that of blank group and model control group. The difference had statistical significance (P＜0.01), Tongsaimai Tablet can increase the blood supply of collateral circulation, lessen muscle fiber atrophy. The effect of Yongsaimai Tablet high dose group was the most obvious.Conclusion: Tongsaimai Tablet can promote blood vessel neogenesis of ischemia limbs, can promote collateral circulation Establishment and improve ischemia symptom. So this study provided theory basis for Tongsaimai Tablet clinical application to treat ischemia disease.
