Studies On Standardized Extract Of Schisandra Chinensis Leaves

For the purpose to provide proof for the medicine use and further exploitation, this thesis studied on the leaves of Schisandra chinensis in the point of pharmacognosy and preparative technology and quality control methods of standardized extract.1. Pharmakognosie of leaves Schisandra chinensisThis thesis studies the character, microstructure, TLC of the leaves of Schisandra chinensis from pharmacognosy, assaies the content of the extract, ash and total lignans in the leaves. The content of total lignans from Schisandra chinensis leaves collected in different seasons is also compared.2. Extraction technology of standardized extract of Schisandra chinensis leavesComparing three methods including reflux, ultrasonic extraction and maceration, the best extraction method were determined. The optimum extraction routine was confirmed by the comparison of two technical routines. The optimum water decoction technology was screened according to the factors affected the extraction such as the quantity of water added, reflux time and extraction times with the content of total lignans as index by orthogonal experiments. The optimum ethanol extraction technology was screened according to the factors that affected the extraction such as the quantity and concentration of ethanol, reflux time and extraction times with the content of total lignans as index by orthogonal experiments.3. Purification and enrichment of standardized extract of Schisandra chinensis leavesThe type of macro porous adsorption resin was selected by orthogonal experiments and single factor. Furthermore, the best purification technology of standardized extract was determined after the parameters including elution solvent, initial sample concentration, diameter to height ratio and flow rate.4. Quality control of standardized extract of Schisandra chinensis leavesThe content of total lignans and schisandrol A of standardized extract were determined by ultraviolet spectrophotometry and HPLC, respectively. According to the methods included in Chinese Pharmacopoeia（2005th）, the ash, heavy metal and water content were examined. Finally, 8 chromatographic peaks in standardized extract were identified as schisandrol A, schisandrol B, schisandrinB, schisandrinC, Gomisin G, Gomisin L₁, Gomisin M₁ by HPLC.
