The Pharmacodynamics Research Of HRG On Climacteric Syndrome

Hongrengui capsule is a fomula to remedy Climacteric syndrome ,which was picked up from components of honghua （Carthamus tinctorius）, renshen（Panax ginseng）, danggui （Angelica sinensis）,gouqizi （Lycuim barbarum）,suanzaoren（Zizyphus spinosa）, fuling（Poria cocos） and yinyanghuo（Epimedium brevicornum）through modern science and technology. Two experimental animal models, MP model and OVX model, were used to study the effects of HRG on Climacteric syndrome and its mechanisms.It was found that HRG can make OVX rats in oestrum, increased uterus and adrenal index and markedly increased the content of serum E₂ in OVX rats. HRG gradually made the irregular estrous cycle of MP mice regular, increased uterus、ovary and adrenal index of MP mice. So we can proved that HRG can regulate reproduction endocrine effect. HRG can evidently lighten thymus and spleen atrophy degree of MP mice and OVX rats, which indicated that it can improve immunity. It was showed that HRG can decrease the content of serum MDA and increase the level of SOD in MP mice, from which we can infer that HRG has antioxidation. It was showed that the contents of serum T-CHO and TG decreased and the content of serum HDL-C increased in OVX rats because of HRG. From which we can infer that HRG can inhibit atherogenesis. HRG increased the contents of serum calcium and serum phosphorus and product of serum calcium and phosphorus, decreased the level of serum AKP in OVX rats, which indicated that HRG has good effects on osteoporosis. The fact that HRG improved learning and memorizing ability of passive avoiding harmful things in MP mice indicated that HRG can improve learning and memorizing ability. HRG reduced the less index and fat index in MP mice and OVX rats, which showed that HRG can inhibit climacteric spontaneous obesity. It was found that HRG increased the grip time, the hypoxia tolerance when in ordinary pressure and the survival time in water of MP mice. We may make conclusions that HRG can improve the function of resistance against exhaustion and stress. HRG can restrain the autonomic activity and shorten the hypnogenetic latency period, extend the sleeping time induced by pentobarbital sodium, from which we can infer that HRG has the sedation effects. Hypothalamic monoamine neurotransmitters （NE, DA, 5-HT and 5-HIAA） contents were determined by high performance liquid chromatography and electrochemical detector. HRG can reduce the contents of hypothalamic 5-HT and 5-HIAA, which were increased in models and it can increase reduced NE and DA of hypothalamus in models. It also can let the ratio of 5-HT to NE decrease nearly to the normal level group, which indicated that HRG can regulate the function of nerve system.Summarizing all the experimental results, HRG produce effects on Climacteric syndrome by means of regulating the function of plant nerve and reproduction endocrine and body immunity. It can increase the level of female hormone secretion, antioxidation, inhibit atherogenesis, defend osteoporosis ,improve the metabolism of free radicals efficiently etc, slower the senility of middle age and old women.
