Backgroup and Objective:P53gene is one of the most important tumorsuppressor gene,also is the most common mutant gene of a variety of humantumors, including esophageal cancer. Wild-type p53function having anti-cellproliferative, p53gene mutation can damage their DNA binding andtranscription factor function,its normal function of controls the cell cycle andcell proliferation was inhibited.P53as a tumor suppressor gene, by regulatingDNA repair, cell cycle regulation, inhibition of angiogenesis and induction ofapoptosis and other mechanisms play an important role in the growth,development and differentiation of cells in the body’s tissues. Esophageal canceris one of the common digestive in China,numerous studies confirm that,p53issignificance for esophageal cancer mechanism, early diagnosis, treatment andevaluation of prognostic.This study to explore the relationship between geneP53expression and the concurrent radiotherapy and chemotherapy forEsophageal, provide a reference for predicting the effect of esophagealcarcinoma treatment.Methods:Using hospital-based case-control study and detect the expression of p53mRNA in peripheral blood leukocytes of50cases of esophageal carcinomapatients by RT-PCR(reverse transcription-PCR) method.Patients are to beenrolled in a total of60-70Gy radiotherapy and two cycles of conventionalsynchronous DF (fluorouracil and cisplatin) chemotherapy. Analysis therelationship between p53gene expression and radiotherapy with concurrentchemotherapy. SPSS17.0statistical analysis software for data analysis,t testwas used to compare measurement data,chi-square test was used to compare thecount data,statistical tests were two-sided test, test level α=0.05.Results:(1)In positive group the P53mRNArelative expression of peripheral bloodleukocytes is0.43±0.14,in negative group the P53mRNA relative expressionof peripheral blood leukocytes is0.09±0.02,the difference was statisticallysignificant (p <0.05).(2)12cases in the group of P53-positive patients’therapy is effective,efficiency of66.7%,32cases in the group of P53-negative patients’ therapy iseffective, Efficiency of96.9%.The efficacy of the50cases of esophagealcarcinoma patients who took the concurrent radiotherapy and chemotherapy ishighly correlated with p53expression in peripheral blood. Efficacy of P53-negative patients was significantly better than P53-positive patients(p<0.05).With further performed analysis,the results showed: in age, radiation dose, withor without chemotherapy before treatment, esophageal cancer family history,smoking, drinking there the efficacy of chemotherapy were no statisticallysignificant differences (p>0.05).Conclusion:P53gene overexpression of esophageal carcinoma cases occur athigh frequencies,over-expression of p53gene or not may influence thesensitivity of tumor cells to radiotherapy and chemotherapy.Therefore, to detect the expression of p53gene may help to determine the response of esophagealcarcinoma to the radiotherapy and chemotherapy, and to determine theprognosis of patients. It has a certain reference value for clinical cancer therapy.
