Objective: To observe the effect of Longzhi Tang (Since the proposed name) to the Vascular Endothelial Growth Factor (VEGF) and its receptor Fetal Liver Kinase1(Flk-1) mRNA expressed. The two genes come from the focal cerebral ischemia SD rat models’peri-infarction tissue. Primary discuss the mechanism of how Longzhi Tang affects the recovery stage of cerebral infarction, and provide the experimental basis for clinical application.Methods:Choose120male healthy SD Rats, and made the model of focal cerebral ischemia rats by the suture occluded method, than randomly divided into Experimental Group, Control Group, and Blank Group,40rats per group. The model rats need intragastric administration twice a day after2h of the operation. Give Longzhi Tang to the Experimental Group, Buyang Huanwu decoction to the Control Group, and clean water to the Blank Group. Randomly execute10rats in each group after the treatment in day3, day7, day14and day28. Collect the peri-infarction of brain by autopsy. Through the RT-PCR method test the expression of the VEGF and its receptor Flk-1mRNA in brain tissue. Results:The expression of VEGF mRNA in the above three groups were significantly decreased from day3. The decline rates of the expression of VEGF at the Experimental Group were quicker than the Control Group and the Blank Group in3d-7d (Slope a,-0.26<-0.17=-0.17), but the decline rates were slow down in7d-14d and14d-28d (-0.13>-0.14>-0.18;-0.1>-0.17>-0.11). The expression of Flk-1mRNA began to increased temporary in the three groups of rats in3d-7d,7d later started to reduce gradually. The increased rates of the expression of Flk-1mRNA at Experimental Group were quicker than the Control Group and the Blank Group in3d-7d (0.17>0.13=0.13), but the decline rates were slow down in7d-14d and14d-28d (-0.11>-0.12>-0.13;-0.21>-0.26>-0.37). Through the correlation analysis, the expression of VEGF and Flk-1mRNA in the three groups has a correlation with time, and the correlation between time and the expression of VEGF&Flk-1mRNA in the Experimental Group were higher than the Control Group and the Blank Group (Correlation coefficients r,0.718>0.578>0.562;0.617>0.536>0.529).Conclusion:Longzhi Tang can promote the peri-infarction tissue of angiogenesis may through the affect of VEGF and its receptor Flk-1genes; obtain the objective of brain rehabilitation.
