Study On The Relationship Between VCAM-1 Gene Polymorphism With Essential Hypertension

Abstract/Summary:
Objective:Investigate the association between VCAM-1 single nucleotide polymorphism and essential hypertension susceptibility, to provide scientific evidence for the prevention and treatment of essential hypertension.Methods:A case-control study in groups was performed.407 essential hypertension patients (cases) and 677 normal residents (controls) were recruited in Nanning community residents, and their information of demographic data and risk factors were collected by a unified questionnaire. A 3 ml blood sample was drawn to extract genomic DNA in the method of Phenol-Chloroform. The four SNPs of VCAM-1 gene were genotyped with Matrix-assisted laser desorption ionization time-of-flight mass spectrometry technology (MALDI-TOF-MS). Measurement data were compared with group t test, count data were compared with x 2 test; Multivariable logistic regression model were used to evaluate the association between gene polymorphism and essential hypertension risk. MDR method was used to test the interaction between four SNPs of VCAM-1. Hardy-Weinberg genetic equilibrium (HWE)、LD analysis、haplotype analysis were tested in SHEsis software (http://analysis2.bio-x.cn/SHEsis Main.htm). All tests were two-sided and a P value of <0.05 was considered statistically significant.Results:(1)The general characteristics of the subjects:The distributions of age and alcohol consumption were obviously difference between cases and controls (P< 0.05).We had not found any statistical differences in the distributions of cholesterol(TG), triglycerides(TC) and high density lipoprotein cholesterol(HDL) between cases and controls (P>0.05), respectively.(2)Association between VCAM-1 single nucleotide polymorphism and essential hypertension susceptibility:The genotype frequency distributions of VCAM-14 SNPs were in agreement with the Hardy-Weinberg Equilibrium in controls (P>0.05). There were no statistically differences of the allele frequencies and genotype between cases and controls (P>0.05).After adjusted the factors of age and alcohol consumption, there was no significant association between VCAM-1 gene polymorphism and the risk of essential hypertension (P>0.05).(3) Interaction analysis of VCAM-1 gene polymorphism:Logistic regression models found that VCAM-1 SNPs and environment factors interactions were not statistically significant associated with essential hypertension (P>0.05). The MDR analysis did not observe any significant reaction between VCAM-1 SNPs in relation to essential hypertension (P>0.05).(4) Haplotype analysis of VCAM-1 gene polymorphism:There were 6 haplotypes of VCAM-1 four SNPs and the haplotype C-G-T-A frequency of in case was significant higher than control (P=0.036). The haplotype C-G-T-A was a susceptible factor of essential hypertension (OR=1.230,95%CI=1.013-1.493).Summary:(1) There were no association between VCAM-1 (rs3176867、rs3176875、 rs3917012-. rs3917018) gene polymorphism and essential hypertension.(2) The haplotype C-G-T-A (rs3176867、 rs3176875、rs3917012、rs3917018) of VCAM-1 increases the risk of essential hypertension.
