Enrichedwith chronic obstructive Pulmonary disease Prevention and treatment in Chinesemedicine science content.Method:Method of making model by composite factors, combined with the establishmentof a rat model of chronic obstructive pulmonary disease with phlegm and blood stasissyndrome.Male SD rats were randomly divided into normal control group, Modelgroup,the low-doses groups of Qibai Pingfei Capsule, the high-doses groups of QibaiPingfei Capsule,Fasudil group (n=20for each group).After model establishment, Pul-monary function observed in the ratã€Hemodynamicsã€The changes of pulmonaryvascular morphologyã€ELISA determination of Rho associated coiled coil containingprotein kinase1(Rock-1) and Rho related protein kinase2(Rock-2) levelã€Immunohistochemical detection of Rock-1,Rock-2protein expressionã€Western blotanalysis of pulmonary Rock-1and phosphorylated MYPT-1protein level and Thetranscriptional level of RT-PCR detection of Rho kinase mRNA.Results:(1) The experimental results showed that there was no significant difference of pulmonary function of Qibai pingfei capsule in improving model rats and effect ofFasudil.(2) Laboratory examination showed Qibai pingfei capsule could reduce thelevel of serum Rock-1,Rock-2,PDGF and HIF-1α levels,compared with the modelgroup,P<0.01or P<0.05,there was statistical significance.(3) Pathological examinationshowed Qibai Pingfei capsule can obviously reduce the inflammatory cell infiltrationin lung tissue of rats and the proliferation of smooth muscle cell hypertrophy.(4) QibaiPingfei capsule can decrease the transcription level in lung tissue of rats with Rock-1mRNA and Rock-1and the phosphorylation of MYPT-1protein expression level,compared with model group, P<0.01or P<0.05,there was statistical significance.Conclusion:Involvement of the Rho/Rock pathway in pulmonary vascular remodeling, QibaiPingfei capsule can effectively alleviate the pulmonary vascular remodeling, and itsmechanism may be related with the expression regulation of Rho kinase in serum,lungtissue and pulmonary artery. |
