Objective:To study the expression of IL-5in human nasal polyp tissues, and to investigate the possible relevance between eosinophil counts and IL-5concentration.Methods:The enrolled patients were divided into three groups according to the report of history,nasal endoscope, CT and pathology,laboratory examination:control group(n=13), NP group(n=22), NP with AR group(n=20).which is named as the nasal polyps(NP) group and the health control group. The control group was only with deviation of nasal septum,and part of the middle turbinate hypertrophy.Removed polyp tissues under nasal endoscope and sectioned to save. Eosinophil counts were measured by HE staining, IL-5expression were detected by immunohistochemistry and ELISA.The group differences of eosinophil counts and IL-5expression were analyzed by one-way ANOVA, and relevance between eosinophil counts and IL-5concentration were analyzed by Pearson correlation.Results:1. HE staining:The organizational structure of nasal mucosa in control group was clear and complete, and eosinophil infiltration was rare. The edema of nasal mucosa in NP group and NP with AR group were obviously, and numberous eosinophils infiltrated into nasal mucosa. Eosinophil counts were:control group (1.2±0.4)/HP, NP group(7.4±1.6)/HP, NP with AR group (13.5±2.5)/HP. Eosinophil counts had significantly differences among the three groups(F=180.1, P<0.001).2. Immunohistochemistry staining:The cytoplasm characteristic of IL-5positive staining was brownish yellow or brown granules. The staining of control group was slightly compared to NP group and NP with AR group. The average grey value of IL-5positive staining cells were:control group (177.3±21.1), NP group(124.8±7.4)/HP, NP with AR group (93.3±8.1)/HP. The average grey value of IL-5positive staining cells had significantly differences among the three groups(F=187.3,P<0.001).3. IL-5concentration(ELISA method):IL-5concentration was calculated according to standard curve. The detailed concentration were:control group (1.3±0.2) ng/ml, NP group(2.9±0.3) ng/ml, NP with AR group (4.8±0.5) ng/ml. IL-5concentration had significantly differences among the three groups(F=317.1, P<0.001).4. Correlation between IL-5concentration and eosinophil counts:The relationship between IL-5concentration and eosinophil counts was not relevant significantly(r=0.48, P=0.1). IL-5concentration and eosinophil counts were significantly correlated, both in NP group(r=0.86, P=0.001) and NP with AR group(r=0.84, P=0.001).Conclusion:The eosinophil counts and IL-5concentration of polyp tissues in NP group were increased significantly, and more obviously in NP with AR group. The degree of eosinophil infiltrated into of nasal mucous was closely related to IL-5expression,and IL-5may involved in the pathogenesis of NP.
