Background:Paraquat, it’s trade name is1,1’-dimethy1-4,4’-bipyridinium dichloride.Paraquat inactivates quickly while contacting with earth, having no atmosphere pollution and residue exists.Since paraquat is excellent in weeding and has a poor price, it is widely used in agriculture country. The number of people who attempt to suicide or accidental poisoning is still high because of the eazy chance of exposure to it. After intaked, lung, liver, kidneys, heart, and neural organization would be damaged. Multiple organ of the body function failure is induced. The major target organ is the lung, acute lung injury and advanced pulmonary interstitial fibrosis is the most serious complication. The mechanism is still indefinite, and no special antidotes exist, so the mortality for the study of the mechanism of paraquat poisoning is high. Cordyceps-Astragalus-SalviaMiltiorrhiza is a capsule, contents of beige powder, tastes of a little weeds and bitter. Its content includes Chinese caterpillar fungus, astragalus membranaceus and salvia miltiorrhiza,safflower, zizyphus jujube kernel (Fried). Pharmacological studies confirm that, to Ma Sugi nephritis model for the rabbit and rat animal model of heterogeneous serum type immune complexes nephritis, it can reduce urinary albumin, increase serum albumin, reduce the kidney immune complex deposition. It is widely used in pulmonary fibrosis, pulmonary hypertension, and immune related kidney damage. It can reduce the release of inflammatory factors in acute injuries, improve hemodynamics, raise exercise tolerance and increase life quality. Very few related literature was reported in Cordyceps-Astragalus-SalviaMiltiorrhiza therapy of paraquat.Objective:Our research study the therapy of chongcaoqqishen in treating lung injury and pulmonary interstitial fibrosis induced by paraquat.Method:All90adult Wister male rats were randomly assigned to three groups, the paraquat poisoning group(rats were intragastric administration paraquat50mg/kg body weight once at the beginning); the Cordyceps-Astragalus-SalviaMiltiorrhiza therapy group(rats were given Cordyceps-Astragalus-SalviaMiltiorrhiza90mg/kg body weight intragastric administration half an hour after paraquat was given, then the same dose was given once a day); control group(rats were intragastric administration with physiological saline). At7th,14th,21st,and28th day rats were sacrificed postanesthetic respectively after paraquat exposure, sample of lung tissue, bronchoalveolar lavage (BAL),and venous blood were collected. GSH, SOD, TNF-α, TGF-β,and HYP in plasma, bronchoalveolar lavage (BAL), and the lung homogenates were determined. Optical microscope was performed to examine pathological changes in lung.Result:GSH, SOD of paraquat poisoning group were significantly lower than control group, the therapy group was significantly higher than paraquat poisoning with the same days. TNF-α, TGF-β,and hydroxyproline of paraquat poisoning group were significantly higher than control group, the therapy group was significantly lower than paraquat poisoning with the same days. Under optical microscope, the tissue damage of lung was aggravated, and reduced after Cordyceps-Astragalus-SalviaMiltiorrhiza was administrated.Conclusion:Cordyceps-Astragalus-SalviaMiltiorrhiza can reduce inflammation factor releasing, and relieve lung injury. It has therapeutic effect on lung injury induced by paraquat poisoning.
