Experimental Study Of Cordyceps Sinensis Powder In The Treatment Of Acute Lung Injury In Rats

Objective: Research on cordyceps sinensis powder in the treatment of acute paraquat(PQ)poisoning of lung injury in rats,and analysis of the related mechanism.Methods: Choose 210 healthy adult SD rats(male and female half),were randomly divided into A,B,C,D group: group A(healthy controls)35: give A 2.5 ml by intraperitoneal injection of saline,daily to give 5 ml saline lavage;Group B(pure infected group)35: give a PQ intraperitoneal injection of 20 mg/kg,PQ concentration of 2 mg/ml,daily to give 5 ml saline lavage;Treatment group C(from cordyceps sinensis C1,C2 and C3 group,low,medium and high dose of 35)only 105: give a PQ intraperitoneal injection of 20 mg/kg,PQ concentration of 2 mg/ml.Low,medium and high dose daily 0.3 g/kg to give from cordyceps sinensis,1.5 g/kg,5 g/kg,5 ml saline lavage after dilution;Group D(vitamin E treatment group)35: give a PQ intraperitoneal injection of 20 mg/kg,PQ concentration of 2 mg/ml.Give daily vitamin E 10 mg/kg,5 ml saline lavage after dilution.Both groups of rats from building after 1 d,3 d,7 d and 14 d and 21 d,28 d and 35 d observation,observe the general conditions,the colour,breathing,mental state,eating,etc.Infected group,treatment group(low,middle and high dose),vitamin E treatment group respectively in the above five rats from each time node,execution,take blood to return blood specimens,the determination of malondialdehyde(MDA),superoxide dismutase(SOD),glutathione peroxidase(gsh-px),in 7 days,21 days in bronchoalveolar lavage fluid,blood specimens were T lymphocyte subgroup.Blank control group at the same point in time will take 5 rats to determine the above indicators,at the same time,the above groups are in the left lung of rats,using filter paper suck blood,be bronchoalveolar lavage(BALF),medial right lung pathological tissue preparation,for hematoxylin eosin staining(HE),pathological observation under light microscopy.In BALF and peripheral blood T lymphocyte subsets(CD3 +,CD4 +,CD8 +,CD4 + / CD8 +)to the expression of a fluorescent direct notation,flow cytometry instrument test.Application of immunohistochemical detection of PQ poisoning with acute lung injury in the lung tissue of alpha 1-antitrypsin(alpha 1-AT)expression.Alpha 1-an immunohistochemical staining positive biopsy with 40 HP objective selected 10 vision observation under positive cell cytoplasm average absorbance(OD),with its relative content reaction staining intensity per unit area.Results:1.The general situation of rats: A group of rats showed good mental state at each time point,mobility,breathing smooth,lively and active,fur luster bright,eat more,difficult to capture;C(middle,high dose group,the group D early spirit is a bit poor,after a shortness of breath,the person that weigh after oral,limbs and tail light purple,purple,a week after treatment of symptoms,eating,and active and normal mental state;Early symptoms than in group C(low dose),high dose group,hair loose,which appear when resting oral,limbs and tail light purple purple,eat less,spirit bad,after treatment of symptoms for a medium and high dose group.Group B after infected mental state is poor,hair loose,unsteady gait,eat less,prostrate,easy to capture,shortness of breath,oral,limbs and tail purple purple obviously.2.The lung tissue injury of C(middle,high dose group compared with group B pathology change obviously,significantly improve lung tissue damage degree,low dose group compared with group B must be improved.Compared with group A,group D pathology change obviously,lung tissue injury was significantly improved.3.In serum MDA,SOD,gsh-px levels: C(low,middle and high dose)compared with group B,group level are better than that of group B(P < 0.01,P < 0.05);Compared with group A,group D levels were significantly better than that of group A(P < 0.05).4.In the peripheral blood T lymphocyte subsets(CD3 +,CD4 +,CD8 +,CD4 + / CD8 +): 1,3 weeks,C1,C2 and C3 group of venous blood CD3 +,CD4 +,CD8 +,CD4 + / CD8 + a rise in group B(P < 0.01,P < 0.05),the bigger the cordyceps sinensis powder treatment dose,the longer the treatment time,CD4 + / CD8 + ratio normalizes.Group D venous blood CD3 +,CD4 +,CD4 + / CD8 + were rising in the B group(P < 0.01,P < 0.05).5.In the BALF of T lymphocyte subsets(CD3 +,CD4 +,CD8 +,CD4 + / CD8 +): 1,3 weeks,C1,C2 and C3 group BALF CD3 +,CD4 +,CD4 + / CD8 + levels increased compared with group B(P < 0.01,P < 0.05),the bigger the cordyceps sinensis powder treatment dose,the longer the treatment time,CD4 + / CD8 + ratio normalizes.Group D BALF of CD3 +,CD4 +,CD4 + / CD8 + a rise in group B(P < 0.01,P < 0.05).6.In the lung tissue of alpha-1 AT: group C compared with group B,group C(low,middle and high dose)in lung tissue of alpha 1-an expression is significantly higher than group B(P < 0.05),group B compared with group D with no statistical significance(P > 0.05).Conclusion:Cordyceps sinensis powder for PQ poisoning caused by acute lung injury(ali)has good treatment effect,can eliminate oxygen free radical,improve the body's resistance to oxidation damage,inhibition of lipid peroxidation,and can promote the alpha 1-an expression,balance antitrypsin and protease level,and improve the level of CD3 +,CD4 +,improve CD4 + / CD8 + ratio,immune regulation,so as to reduce the lung tissue damage caused PQ poisoning.
