Objective: Through in vivo and in vitro experiment to study and analyze theeffect of soothing liver and regulating the qi method in promoted gastricdynamic effect and explore the mechanism from the GASR-PLC-IP3-Ca2+signalpathway.Methods: In vitro experiment①32rats were selected and randomly dividedinto4groups: control group, model1group, model2group andChaihushugansan group (each group n=8). The model1group was establishedby constantly squeezing rat tails for4wks. The latter two groups wereestablished by constantly squeezing rat tails together with increasing hunger orovereating for4wks.②With the same model started from the first day,Chaihushugansan decoction (141%concentration),1.5ml/100g,wasadministered to the Chaihushugansan group and normal saline was administeredto the other groups by using the gavage technique. Open-field test was detected weekly. After four weeks, gastric emptying and intestinal propulsion functionwere detected. In vivo experiment①5%,10%,20%dose of serum containingChaihushugansan,10%dose serum containing motilium and blank serum wereprepared.②Smooth muscle cells (SMC) of gastric antrum were isolated bycollagenase type II digestion method.③We compared the SMC contractileresponse of rat gastric antrum from5%,10%,20%dose serum containingChaihushugansan,10%dose serum containing motilium and blank serum underthe inverted microscope and determined the optimal effective concentration ofserum containing Chaihushugansan.④Each time one type of blockers includinggastrin receptor (GASR) antagonist L-365260、phospholipase C (PLC)inhibitor U-73122and intracellular calcium antagonist TMB-8were added intonormal rat antrum SMC. We prepared the permeability in gastric antrum ofSMC with the inositol triphosphate (IP3) receptor antagonist heparin, added thebest dose of serum containing Chaihushugansan for the effect on contractility.The contractile response on rat gastric antrum SMC was observed to determinethe signal transduction pathways which led to the contraction effect of rat gastricantrum SMC induced by serum containing Chaihushugansan.Results:In vitro experiment①All rats in model groups showed pad batch,margination, move less, aggressive reduction, easy to rage, withered hair. Theabove manifestations in Chaihushugansan group were not so obvious comparedwith those in the model groups. Then, after four weeks, all rats were sacrificedand laparotomized. Gastric mucosa in the control group had no significantflushing, swelling, erosion and other diseases, while the model groups had muchgastric contents and flushing. The above manifestations in Chaihushugansangroup were not so obvious compared with those in model group.②The sugar consumption, the scores for horizontal motion in the open-field test, and therates of gastric emptying and intestinal transit function of all model group wereless than the control group(P <0.05). The sugar consumption、the scores forhorizontal motion of open-field test、gastric emptying and intestinal transitfunction in model2group were less than model1group(P <0.05). The sugarconsumption、the scores for horizontal motion of open-field test、gastricemptying in Chaihushugansan group were more than model2group(P <0.05). The scores for vertical motion of open-field test were in model2groupless than model1group. The scores for vertical motion of open-field test inChaihushugansan group were more than model2group but there were nosignificant difference. In vivo experiment①The contractile responses of ratgastric antrum SMC induced by5%、10%、20%concentrations of serumcontaining Chaihushugansan and10%concentrations serum containingmotilium were more obvious than those in the blank serum(P <0.05). Themost obvious contractile response of rat gastric antrum SMC induced by20%concentrations of serum containing Chaihushugansan. The contractile responseof rat gastric antrum SMC in20%concentrations of serum containingChaihushugansan was not obvious compared with10%serum containingmotilium.②When blank serum、L-365260, U-73122, Heparin and TMB-8were added,20%concentrations of serum containing Chaihushugansanobviously reduced the contractile response of rat gastric antrum SMC(P <0.01).Conclusions: The method of soothing liver and regulating qi had a role topromote the gastric motility of functional dyspepsia rats. Its mechanism mightbe based on the GASR-PLC-IP23-Ca+signal pathway.
