Objective To verify the hypothesis that stimulation of the nucleus basalis of Meynert (NBM) in rats with vascular dementia can alleviate the cognitive impairment, we investigated whether the expression of NGF and Chat in CA1region of hippocampal neurons is increased after stimulation of NBM using immunocytochemistry techniques.Methods We used adult male Sprague-Dawley rats (weight300-400g).Rats were randomized into sham operation group, sham stimulation group and stimulation group. VaD rats were established by bilateral vertebral artery occlusion, we perform chronic stimulation (30min per day,14days) on2-VO rats and examine the spatial learning and memory in Morris water-maze. The three group perform continuous4days place navigation test and the fifth day perform spatical probe experiment. Application Nissl staining and immunohistochemical staining technique to observe changes in hippocampal CA1neurons of rats in difference group.Results In the place navigation test, compared with model group, the stimulation group was shorter following elongation of reperfusion (p<0.05); in the spatical probe experiment, In perform spatical probe experiment, percentage of time quadrant, the number and area by swimming swim platform from the platform quadrant of the stimulation group was the more than sham stimulation group, the difference was statistically significant.Hippocamal CA1regions the number of Nissl bodies was not reduced in sham operation group and stimulation group, but decreased in model group. Compared with model group, in Hippocamal CA1regions the expression of NGF and Chat was obviously increased in stimulation group (p<0.05).Conclusions The result suggests that stimulation of NBM in rats with vascular dementia can alleviate the cognitive impairment, which through increasing the expression of NGF and Chat in CA1region of hippocampal neurons.
