Objective To examine the expressions of Transient receptor potential channel TRPM8and its clinical significance in Human Pancreatic Ductal Adenocarcinoma (PDAC).Methods The tests were collected from Sixth People’s Hospital, Shanghai JiaotongUniversity Hepatobiliary Surgery March2008to March2010of180cases of surgicalresection of pancreatic ductal adenocarcinoma cases,40cases were randomly selectedpathological and clinical data better cases.We determined the expression levels of TRPM8on both steady-state mRNA and protein levels in40PDAC samples by quantitativereal-time PCR and immunohistochemistry, respectively. The correlations between TRPM8expression and various clinicopathological parameters were analyzed.Results The expression of TRPM8on the mRNA level was consistent with that on theprotein level. TRPM8mRNA levels in tumor tissues were significantly higher than in theparatumor tissues (0.0512±0.0498vs0.0008±0.0005; P<0.05). Univariate andmultivariate analyses revealed that TRPM8expression was an independent prognosticfactor for disease-free survival and overall survival. Both2-year disease-free survival ratesand overall survival rates in TRPM8-positive group were significantly lower than innegative group (48vs.18%,58vs.22%). Clinicopathological characteristics evaluationresults: High mRNA expression of TRPM8significantly correlated with thedifferentiation,TNM stage and lymph node metastasis of the tumor(p<0.05). Nosignificant correlation was found between TRPM8mRNA expression and other variablesincluding tumor diameter, patient age and gender (P>0.05).Conclusions Our results suggest that TRPM8expression is up-regulated in PDACand its expression is Closely associated with several clinical and pathological features forPDAC. This suggests that TRPM8may be a important biological indicators and reflectethe development, invasion, metastasis and prognosis of pancreatic ductal adenocarcinoma.
