Objective Tetrastigmatis hemsleyani named Tetrastigma hemsleyanum Diels et Gilg is one plant of Family Vitaceae and Tetrastigma obtectum Genus. Modern pharmacological studies have shown that it has good antitumor activity. However reports of antitumor active ingredient of Tetrastigmatis hemsleyani are rare.And the material basis of antitumor effet is not very clear. Based on this, this paper is intended to be pharmacologically active as a guide, screening the antitumor active fractions of Tetrastigmatis hemsleyani, then separating and purifying the active fractions, thereby screening the active ingredients, and identifying their chemical structures. This will provide some scientific basis on antitumor activity of Tetrastigmatis hemsleyani.Methods All fractions of Tetrastigmatis hemsleyani was got by solvent extraction system. Furthermore the antitumor active fraction was screened by means of MTT in which MB-435s cell was used as test subject.Then some chemical compounds of the above active fraction was gotted by system plant seperation,such as cilica gel column chromatography, Sephadex LH-20chromatography, Low pressure preparative LC chromatography, preparative HPLC chromatography, HPLC, macroporous resin and other Separation and purification methods. Finally antitumor active compounds screened by cell assays (means of MTT) in vitro was found.Results The results of cell assays in vitro was displayed that the fraction of diethyl ether and water ware showed definite antitumor activity.Their IC50ware99.7μg/mL,127.8μg/mL respectively and their efficacy was positively correlated with the concentration of drug. The ethyl ether fraction, one of the antitumor active fraction, was isolated and purified systematically.Then10chemical compounds ware gotted. They are β-sitosterol(D1), Palmitic acid (D2),3,4-Dihydroxybenzoic acid (D3),5-(4-hydroxystyryl) benzene-1,3-diol(D4), Kaempferol (D5), Quercetin (D6),4-Hydroxybenzoic acid (D7), salicylic acid (D8), trans-4-Hydroxycinnamic Acid (D9, Isoquercitrin (D10).2active ingredients were found by screening above chemical compounds. They are5-(4-hydroxystyryl)benzene-1,3-diol(D4), Kaempferol (D5) and their IC50ware92.4μ/mL,294.2μg/mL respectively. The fraction of water,the other of the antitumor active fraction, was isolating by the method of macroporous resin and alcohol precipitation. Afterwards6fractions ware gained. They are ZF1, ZF2, ZF3, ZF4, ZF5, ZF6. It is obtained that3antitumor active fractions owere found by screening the6above fractions. They ware ZF2, ZF3, ZF4.Conclusion6ingredients of10chemical compounds which ware isolated from ethyl ether fraction ware obtained firstly. They ware D2, D3, D4, D7, D9, D10. It is firstly screened that the active ingredients of Tetrastigmatis hemsleyani. And2antitumor active compounds ware gained. The fraction of water of Tetrastigmatis hemsleyani was studyed in vitro. Efficacy of ZF3and ZF4was positively correlated with the concentration of drug. This dissertation has an significant effect on clarifying pharmacodynamic material basis of Tetrastigmatis hemsleyani. It also enriched the research on active ingredients of Tetrastigmatis hemsleyani.
