Objective: Aucklandia lappa Decne. has an effective on promoting qi circulation to relieve pain and the processed Aucklandia lappa Decne. reinforces its effective on checking diarrhea, which were documented in China Pharmacopoeia.It is clear that the change happens on effective of Aucklandia lappa Decne. before and after procedure, however a thrifle of investtigation of chemical variation for processed Aucklandia lappa Decne. was reported, the mechanism of processing Aucklandia lappa Decne. is still indefinite. So in present paper, the processing principle of Aucklandia lappa Decne. was illuminated on the basis of the chemical composition of Aucklandia lappa Decne., the analysis of the variation of chemical constituents before and after processing, the comparison of pharmacological action of different polarity of Aucklandia lappa Decne. and the dissertion of rule of chemical compositions of Aucklandia lappa Decne. before and after procedure summarized from its processed product, in addition the dependability of the variation of chemical constituents and the change of pharmacological action. On more profound aspect, provides scientific evidence of optimizing procedure of roasted Aucklandia lappa Decne. and establishes the quality criteria of processed product. In conclusion this investigation could be used to indicats clinic medication and confirm the safty and efficacy of the processed product in clinical.Material and method:Material: the dried root of Aucklandia lappa Decne. purchaced from country-wide drug store.Method:1. The chemical compositions were separated using chromatographic technique of silica gel, sephadex-20, HPLC and their structures were determined by physico-chemical property and spectra of 1H-NMR, 13C-NMR and MS.2. The comparation of chemical constituents of Aucklandia lappa Decne. and its processed product was analyzed by UV, TLC and HPLC. The result indicated that the chemical compounds of Aucklandia lappa Decne. influenced significantly by procedure, and the variation of chemical composition of Aucklandia lappa Decne. before and after procedure was regularity.3. The pharmacological mould of splenic asthenia of mouse was serving to investigate the progradation rate of intestina parva and emptying rate of stomach, the correlation of variation of chemical constituents and the effective of Aucklandia lappa Decne. before and after procedure by comparing the pharmacological action of different polarity of Aucklandia lappa Decne. and its processed product.Result:1. 14 chemical compounds were separated and identified as dehydrocostus lactone(1), costun olide(2), 3β-acetoxy-9(11)-baccharene(3), 3-filicanone(4), chrysophanol(5),α-costol(6), (-)-e lema-1,3-11(13)-trien-12-ol(7), caryophyllene oxide(8),α-ionone-3-ol(9),β-ionone-4-keto (10),α-ionone-6-ol(11), dihydro-α-ionone(12), (E)-6,10-dimethyll-9-methylidene-undec-5-en -2-one(13), (E)-geranylactone(14), including chrysophanol,3-filicanone,α-ionone-3-ol,β-ion one-4-keto,α-ionone-6-ol was separated from Aucklandia lappa Decne. for the first time.2. The result of influenced chemical constituents of Aucklandia lappa Decne. in procedure indicated that the both change of quality and quantity happened on Aucklandia lappa Decne. before and after procedure. On the basis of the variation of the content of costunolide and dehydrocostus lactone was marked and the content between this compound was regularity, the modern processing parameter was established that the temperature of procedure was 160~180℃, the time was 15 minutes, the ratio of bran and drug was 1:2.3. Low dose water-solution and volatile oil of rude Aucklandia lappa Decne. inhibit gastric emptying of mouse, promote small bowel peristalsis of mouse; High dose water-solution of rude Aucklandia lappa Decne. inhibit gastric emptying of mouse markedly,have the trend of inhibiting small bowel peristalsis, high dose volatile oil have the trend of promoting gastric emptying and hasten small bowel peristalsis; Low and high dose water-solution of the processed Aucklandia lappa Decne. both show the trend of promoting gastric emptying of mouse and the effective of promoting small bowel peristalsis, low and high dose volatile oil both show the trend of inhibiting gastric emptying and small bowel peristalsis.Conclusion:1. The sesquiterpene is main chemical compositions of Aucklandia lappa Decne., and the dehydrocostus lactone and costunolide are major active constituents of sesquiterpene with an effective on loosing smooth muscle, spasmolysis, cholagogue, and etc. So the dehydrocostus lactone and costunolide could serve as indicatrix of quality control of Aucklandia lappa Decne..2. The comparition of chemical constituents of Aucklandia lappa Decne. before and after procedure is analyzed by UV, HPLC and TLC for the first time. The role of accessories is investigated during procedure on the basis of the chemical compositions of accessories before and after procedure for the first time.3. The thesis provides the basis of procedure of Aucklandia lappa Decne., viewing on the standpoint of influence that the parameters of different procedure impact on dehydrocostus lactone and costunolide, active constituents, of Aucklandia lappa Decne.;summarizes regularity of content of dehydrocostus and costunolide of Aucklandia lappa Decne. before and after procedure,settling material foundation that the dependability of efficacy and chemical compositions is approached.4. The pharmacology is researched, the different dosage and polar part serving as investigative object, its result is the same as the theory that Aucklandia lappa Decne. has an effective on promoting qi circulation to relieve pain, invigorating stomach and promoting digestion; its processed product has an effective on checking diarrhea,and then volatile oil is determined as active part.5. On the basis of the result of changed regularity of chemical compounds of Aucklandia lappa Decne. before and after procedure and pharmacological experiment, dehydrocostus lactone and costunolide are infered that they are the major active constituents and the material foundation of the variation happens on founction after procedure, which is the consequence of two-way interaction.
