Objective:To compare the bone defect repair effect between thrombinpeptide (TP508) composite artificial bone graft and simple artificial bonegraft,to explore the repair effection of TP508composite artificial bone onrabbit radius large segmental bone defect and to provide an initialexperimental basis for clinical application, the animal model of rabbits radiuslarge segmental defect was established.Method:42New Zealand white rabbits were randomly selected toestablish bilateral radial1.5cm bone-periosteum defect animal models.Experimental grouping:randomly select36rabbits,choose the right sideas the experimental side which was implanted into hydroxyapatite artificialbone material and injected into TP508in bone defect region.The left side wascontrol group, which was implanted hydroxyapatite artificial bone materialonly.The remaining6rabbits were used as the sham group and bilateral bonedefect region which were not implanted any substance.Rabbits were killed forgross observation、X-ray examination and hematoxylin-eosin staining atweeks4、8and12after bone defect repair. The Lane-Sandhu score standardwas used for X-rays and histological scores,the Image-Pro Plus imageanalysis software was used for histomorphometry analysis and the SPSSstatistical software was used for the statistical analysis of the results.Results:The scores of X-rays and histological and the percentage ofnewly formed bone in experimental group is higher than its in control groupafter bone defect repair at4、8and12weeks and the difference was statistically significant (P <0.05).The bone defect region of sham group didnot form bone connection.The scores of X-rays and histological in shamgroup is lower than its in experimental group and control group,the differencewas statistically significant (P <0.05).Conclusions:1. Excised rabbit radial1.5cm length can be used as animal models toestablish large segmental bone defects.2.TP508can promote the healing of bone defects, and the compositionof TP508and artificial bone have an good repair effection on rabbits radiuslarge segmental bone defects.
