| Objective: To investigate the early changes of Estrogen receptor alpha(ERα),Estrogenreceptor beta(ERβ), Glucose transporter-1(GLUT-1) and Proliferation cell nuclear antigen(PCNA),Nuclear-associated antigen(ki-67) in the tumor of nude mice transplanted humanbreast cancer cell line MCF-7and Bcap37after Tamoxifen(TAM)treatment.Methods: Tumor-bearing nude mouse models of human breast cancer cell lineMCF-7and Bcap37were established.The breast tumor-bearing nude mouse were randomlydivided into treatment group and control group.The treatment group received tamoxifen10mg.kg-1gastric gavage, once a day and3consecutive days, mice were killed andremove the tumor.The expression of ERαã€ERβã€GLUT-1and Proliferation indexes weredetected by Pathological examination HE(Hematoxylin and eosin staining) andimmunohistochemical SP method. The results of ERα and ERβ immunohistochemistrywere analyzed using semi quantitative method and the results of GLUT-1were analyzedaccording to cell staining intensity;PCNA and Ki-67were scored according to thepercentage of positive cells. The scores of the treatment group and the control groupwere compared.Results:1.The scores of the control group and the treatment group in the MCF-7cell breasttumor-bearing nude mouse were ER α:11.00±0.71and9.60±1.14,GLUT-1:2.72±0.16and2.48±0.13,PCNA:70.80±5.26and61.40±3.91, Ki-67:26.20±2.59and19.16±1.82.The scores reduced and the differences were statistically significance (P <0.05).Thescores of ERβ were10.80±0.84and11.20±0.84,and the difference was no statisticalsignificance(p>0.05).2.The scores of the control group and the treatment group in the Bcap37cell breasttumor-bearing nude mouse were ER α:10.75±0.96and9.20±0.84,PCNA:93.75±2.22and 89.60±1.14, Ki-67:88.80±2.59and82.25±0.50.The scores reduced and the difference wasstatistically significance (P <0.05).The scores of the control group and the treatmentgroup of GLUT-1,ER β were2.68±0.96and2.38±0.24,11.25±0.50and11.20±0.84.Thedifference was no statistical significance(p>0.05).3.The positive rate of proliferation index PCNA and Ki67in the tumor of Bcap37cellhuman breast tumor-bearing nude mouse increased significantly than in the MCF-7cell,and obvious vascular invasion in the tumor of Bcap37cell was observed.Conclusion:1.ERα,GLUT-1,PCNA and Ki-67expression of MCF-7cell decreased after3days ofTAM treatment.2.ERα,PCNA and Ki-67expression of Bcap37cell decreased after3days of TAMtreatment.3.The ability of proliferation and invasion of Bcap37cell were stronger than MCF-7cell. |
