Background:Our country has entered the aging society, with the progress ofthe aging population, he prevalence of hypertension in the elderly population willincrease year by year. Hypertension is a leading risk factor of cardiovascular andcerebrovascular disease,is one of the main reasons of death and disability in theelderly population. in addition to the genes, diet and other environmental factors,Studies have shown that the pathogenesis of hypertension also closely related to theintestinal flora. Human gastrointestinal colonization with a huge number of microbessystem. intestinal bacteria metabolism have irreplaceable nutritionand immuneregulation.But microbial system so huge bring potential pathogenic effects can notbe ignored.The experimental and human studies have confirmed that the animal haschanged the structure of intestinal flora in hypertensive patients, which may be causedby factors of hypertension. At present, the interaction of intestinal microorganismsand human health differences still exist, because they affect the mechanism of humanmetabolism is not clear. Studies have found that, the Firmicutes/intestinal florain Bacteroidetes proportion changes and lipid metabolism and hypertension andother cardiovascular disease are closely related, provides a new direction for the studyof the hypertension, worthy of further study people.Objective:This study from the perspective of the intestinal flora, using real-timequantitative PCR to compare the elderly hypertension and healthy elderly personsEubacterium rectal、Bacteroides thetaiotaomicron、Bifidobacterium genus number ofdifferences. and analyzes the rectum Eubacterium, polymorphous Bacteroides,Bifidobacterium genus correlation with serum lipids, inflammatory markers between,providing new clues to the pathogenesis of hypertension, seeking new targets forprevention and treatment of hypertension. Methods:Selected in2013July-2014January in the First Hospital of JilinUniversity of cadre ward hospital and medical center in elderly patients withhypertension in30, meanwhile30healthy controls. Gender、age、BMI match and theexclusion of secondary other endocrine diseases other cardiovascular diseaseshypertension, coronary heart disease, cancer, liver and other organs of serious damage,diabetes, antibiotics during January and probiotics, nearly four weeks suffering fromdiarrhea or other gastrointestinal diseases. Each experimenter collected fasting bloodsamples, blood、blood biochemistry、C-reactive protein、homocysteine concentrationswere measured. Collection of each object of study on fecal samples, after repeatedcentrifugation to remove impurities, washed, get cell precipitation, extraction kit withbacterial genomic DNA extracted total bacterial DNA. According to the test target-specific primers16SrRNA bacterial gene sequences, using extracted genome DNAas template, amplified by real-time fluorescence quantitative PCR reaction, after thereaction is completed based on the PCR software, calculate fecal sample copynumber bacterial gene, as a quantitative results. The quantitative results intoSPSS17.0statistical software for analysis, Applied between two independentsamples t test to compare group differences. Pearson correlation analysis with rectalEubacterium multiforme Bacteroides, Bifidobacterium copy number index and lipidmetabolism in patients with hypertension, inflammation index correlation. With P<0.05was considered statistically significant.Results:Using two independent samples t test of quantitative PCR results werestatistically analyzed. Eubacterium rectal content is higher in elderly hypertensivegroup (P <0.05); the control content of multiforme Bacteroides and Bifidobacteriumgenus were higher than in elderly hypertensive group (P <0.05). Pearson correlationanalysis showed no correlation was found between the bacteria、lipids and markers ofinflammation in hypertensive patients. Conclusion:This study of the rectum Eubacterium multiforme Bacteroides,Bifidobacterium genus quantitative analysis and comparison, can initially considered:elderly hypertensive patients compared with controls does exist an increase in thenumber of colorectal Eubacterium. Elderly hypertensive patients compared withcontrols exist multiforme Bacteroides, Bifidobacterium genus reduce the number.This is a new research direction of intestinal flora in patients with hypertension.
