Select candidate flies which have impact to total sleep time for repetitive studies by the above methods, to ensure the accuracy of experimental results.Results: In the screening of158chromosome number2targeted gene deletion flies, we got10candidate gene deletion strain flies which have significant differences in sleep time compared with wild-type flies. The sleep time of7517and7837strain flies were significantly decrease (P<0.001) when compare with wild-type CS strain flies, while sleep time of7822,7841, and7797strains significantly longer than wild-type CS strain flies (P<0.001). In female flies, the7787and7495strain increase in sleep time by significance(P<0.001). The7830,7786, and7498strain flies, lengthen sleep time in female, while shorten sleep time in male by significance (P<0.001).Conclusion:1) Df(2L)Exel6034and Df(2L)Exel7069gene fragments correlated with total sleep time of male and female flies.2) Df(2L)Exel8028, Df(2L)Exel9033, and Df(2L)Exel7023gene fragment related to shorten total sleep time of male and female flies.3) Df(2L)Exel7016and Df(2L)Exel6009’ gene fragment reduces total sleep time in female flies, but not in male.4) Df(2L)Exel8034, Df(2L)Exel8008, and Df(2L)Exel6012gene fragment decreases total sleep time in female but increases total sleep time in male flies.
