Research The Influencea Bout Expression Of Gene And Ovarian Repairon On Autoimmune POF Model Mice By Traditional Chinese Medicine Of Bushenhuoxue

Object:Mice were used as experimental subjects to observe the effects about expression of AMH, BMP-15mRNA on premature ovarian failure model mice and investigateing molecular mechanisms of premature ovarian failure by Bushenhuoxue prescription.The ultimate aim is providing experimental basis for clinical application and further development of new drugsMethod:There are66mice, randomly selected11female balb/c mice from all,which are gavaged with NS.The remaining mice were immunized with mouse zona pellucida3(ZP3) to establishe model of premature ovarian failure.The successful model mice were randomly divided into western medicine group, model control group, bushenhuoxue high, medium and low dose group,each group gavaging with progynova, saline, high, medium and low concentrations of liquid of bushenhuoxue.Each mouse successively gavage6days and rest one day,0.3ml/天,which continue for four weeks.After the end of the experiment,we need measurement the weight of mice,blood from eyeball,kill mice by cervical dislocation. Some specimens are used for observing ovarian morphologic changes; some for detecting seruming levels of sex hormones such as E2、FSH、LH by means of ELISA, some to detect the expression level of AMH mRNA, BMP-15mRNA of ovarian tissue by RT-PCR method.Results:(1) The serum level of FSH、LH:Compared with the control group,the level about FSH and LH on each group of the chinese medicine prescription were significantly lower than those in the control group (P <0.01).(2) The serum level of E2:Compared with the control group,the level of E2about the other groups were significantly higher (P<0.01); Compared with western medicine group, the value of high-dose group is slightly higher, but don’t have statistically significant (P>0.05).(3) General situation of mice:Compared with the control group, the weight of western medicine group gain (P<0.01).the mental state of model control group mice are spiritless,however the remaining mice in each group gradually improved. Comparison with other groups,high dose group of chinese medicine and western medicine group were the best performing.(4) ovarian tissue morphology:the change of ovarian tissue morphology between high dose group of chinese medicine and western medicine group are similarlly,can visible all stages of follicle and preovulatory follicles increased,atretic follicles decrease compared with the model group.(5) AMHmRNA:Compared with the model group:the AMHmRNA expression on each dose group of chinese medicine were increased (P<0.05); Compared with western medicine group, mean value of high dose group of chinese medicine is higher (P>0.05), but don’t have statistically significant。(6) BMP-15mRNA:Compared with the model group:the expression about AMHmRNA on high and medium dose group of chinese medicine were increased;Compared with western medicine,the mean value of high dose group of chinese medicine is higer(P<0.01)。Conclusion:1.Bushenhuoxue prescription of traditional chinese medicine can promote follicular growth and ovarian restoring,improve endocrine and ovarian function.the effect of high dose of chinese medicine is best,which is better than western medicine2.the mechanism of prescription of bushenhuoxue treatmenting premature ovarian failure:may improve the expression of AMH、BMP-15mRNA on ovary,promote early follicular hyperplasia and development,improve ovarian reserve,which can inhibite premature ovarian failure.
