| Suaeda Salsa, an herb halophyte belonging to chenopodiaceae, is rich nutritious such as polyunsaturated fatty acids, vitamins and trace elements. From some research in recent years, it has been found that Suaeda Salsa took action on antioxidant, antiatheroscloresis, immunoregulation and anti-inflammatory.In this research, the antioxidant activity of alcohol extraction of Suaeda Salsa was studied and the D-gal aging mice and Drosophila melanogaster were chosen to evaluate the antioxidant effect in vivo. Different polarities of organic solvents were employed to separate from the extract by liquids-liquid extraction. And various antioxidant evaluation systems were chosen to comprehensively evaluate the antioxidant activities in the resulting fraction. Further more, the protection of Suaeda Salsa in D-gal aging mice melanogaster and its mechanism were also investigated, which provided the theoretical evidence for the development of relative products.The results of antioxidant experiments in vitro showed that alcohol extraction of Suaeda Salsa had some antioxidant and radical-scavenging activity. DPPH free radical-scavenging rate of alcohol extraction approached to BHT. Under the concentration of0.1mg mL-1, the inhibition to liposome peroxidation was achieved to74.67%.To investigate the anti-aging ability of alcohol extraction, the D-gal induced mouse aging model was employed.The results showed that high dose of alcohol extraction of Suaeda Salsa (800mg/kg·bw) could obviously improve the learning-memory abilities and acclimatization in behavior test, and meanwhile increase spleen index of D-gal induced aging model mice. It also could reduce MDA content in serum, increase SOD and GSH-Px activity and enhance the antioxidant ability of liver. Middle dose injection (400mg/kg-bw) has the similar action with high dose. Additionally, the ratio of TAOC/MDA in high dose on liver was obviously higher than that in model group.We also investigate the effect of Suaeda Salsa alcohol extraction on Drosophila life span and related indicators. The results showed that each dose of ethanol extraction could prolong the life span and maximal life span of Drosophila significantly, and simultaneously increase CAT and CuZn-SOD activity in Drosophila body.17mg/mL dose of alcohol extraction has showed the most obvious effect in reducing the content·of MDA.Various solvent with different polarity (petroleum ether fraction, ethyl acetate fraction, n-butanol fraction and aqueous fraction) were employed to extract the alcohol extraction of Suaeda Salsa. The antioxidant and free radical-scavenging capacity of the subtractions were determined. The results showed that the decreasing order of DPPH radical-scavenging activity and reducing power among the extractions was n-Butanol fraction> ethyl acetate fraction> aqueous fraction> petroleum ether fraction. The ethyl acetate fraction and n-Butanol fraction can inhibit the lipid peroxidatic reaction of hepatic tissues homogenate in mice and the hemolysis of erythrocyte. The hemolysis of erythrocyte inhibition rate of ethyl acetate fraction was higher than this of other fractions.The D-gal mice were chosen to evaluate the effects of n-butanol and ethyl acetate fractions to the antioxidant system in vivo. The ethyl acetate fraction was better than the n-butanol fraction in reducing MDA in serum and enhancing SOD and CAT activity. The n-butanol fraction was better than ethyl acetate fraction in increasing GSH-Px activity and total antioxidant capacity. Compared with ethyl acetate fraction, the n-butanol fraction was more important to liver oxidation index. Both of them had little role on MDA and TAOC in brain. |
