Rutaceae citrus was full of treasure, All parts of the citrus can be used as medicine. Eaten fresh and also can be processed, such as juice drinks, canned and other processed products. Citrus has a wide area of China’s citrus cultivation, production, and the fruit can become not only citrus processing as raw materials, also is the fruit that people likes. The effectiveness of the composition of the citrus fruit is mainly concentrated in flavonoids, alkaloids and volatile oil and others. In recent years, the research of citrus fruit Limonin was gradually in-depth study. The limonin exist in citrus Rutaceae and Meliaceae plant and it also has some biological activity, just like:anti-cancer, antibacterial, analgesic and anti-anxiety. So we take,19different kinds of citrus in Hunan Province as test material, analysing contents of citrus fruit limonin in the growth and development.The main findings as follows:Creating a high performance liquid chromatography detection method of Limonin. Chromatographic conditions as follows:WeLchrom C18(4.6×250mm,5μm), VAcetonitrile:Vwater=45:55, Velocity of flow1.0mL/min, column temperature30℃, detection wavelength210nm. Through investigation, this method has a good linear relationship between Limonin, R2=0.9998, The average recoveries ranged from92.84%to99.71%. The precision is0.69%. This method also simple and fast for citrus fruit Limonin quantitative analysis.To established HPLC analysis method before and after drying, the seeds of different varieties of citrus fruit peel, pulp parts Limonin content analysis. The results showed that various parts of the different types of mature citrus fruit (seeds, peel, pulp) Limonin content there are significant differences.Research on changes of limonin content in the four citrus fruits including Juhuaxinyou, Xupuchangxing Sweet Orange, Xiangxi Ponkan, and Zhangjiajie Jiujiu, during growth and development. The results show that,4varieties of citrus fruit in the process of growth and development Limonin first increased and then decreased gradually to become stable trend. Among them, the Juhuaxinyou Limonin has a particularly rise downward trend evident.4citrus fruit after flowering120d its Limonin content reaches its maximum value.By single factor experiment and orthogonal experiment to determine the optimum extraction conditions of citrus fruit Limonin. That is, with85%ethanol to the solid-liquid ratio1:30amount extracted25min at55℃, has the highest yield of extract Limonin. Comparison adsorption amount of the resin, the adsorption rate, rate and resolution among D101, AB-8, HPD450, HPD600, DM130,polyamide30-60, polyamide60-100, we selected D101as the limonin purification resin, the adsorption rate as88.53%, a resolution rate as93.47%. D101macroporous Limonin process factors:the sample flow rate of1ml-min", the concentration of the sample to0.5mg·min-1elution flow rate2ml·min-1, eluting alcohol concentration of80%ethanol,the amount of240ml.
