Objective:To compare the structural characteristics of the leukocyte-and platelet-rich fibrin (L-PRF) gels prepared by different centrifugations, recovery rate of the white blood cells and platelet,and release amount and rule of the growth factors in three different L-PRF gels,deepen to understand the biological characteristics of the L-PRF,so as to provide experimental basis for animal experiments and clinical application.Methods:L-PRFs were prepared through three centrifugation methods. The first is centrifugation at2500rpm/min for10minutes, method two at3000rpm/min for10minutes,and the third at4000rpm/min for10minutes.Microstructure and ultrastructure were compared and analyzed by light microscopy and scanning electron microscope (SEM) observation.Complete blood count (CBC) and elisa were used to detect white blood cell and platelet concentration,the release amount of platelet-derived growth factor (PDGF-AB) and transforming growth factor-betal (TGF-betal) in three kinds of L-PRF.Results:(1) All3centrifugations can be prepared L-PRF gels.The main structure of L-PRF is a three-dimensional network structure of fibrin,platelets and white blood cells are mainly concentrated in the red colored portion of the L-PRF gels by microstructure and ultrastructure observation.(2) The amount of platelet and white blood cell in L-PRF gels were significantly higher than that of whole blood.The platelet and white blood cell recovery were higher in2500rpm/min and3000rpm/min,the difference to4000rpm/min was statistically significant (P<0.05),but the difference of2500rpm/min and3000rpm/min was no significant (P>0.05).(3) It has almost same regularity of L-PRF membranes in vitro release TGF-betal,PDGF-AB prepared through three centrifugation methods.The release of TGF-betal,PDGF-AB in L-PRF membranes prepared by2500rpm/min and3000rpm/min is higher than that of4000rpm/min (P<0.05),but the difference between2500rpm/min and3000rpm/min was not statistically significant (P>0.05).Conclusions:The L-PRF gels can be prepared using the three centrifugal method,and cell distribution and fiber structure of the three kinds of L-PRF gel had not obvious difference.The white blood cells and platelets recovery,total release quantity of TGF-beta1,PDGF-AB in L-PRF gel prepared with2500rpm/min and3000rpm/min were higher than that of4000rpm/min,but there is no difference between2500rpm/min and3000rpm/min.24Figures,10Tables,64References...
