Autologous fat grafting has one hundred years of history, the soft tissue of the commonly used as filler material, biological compatibility is better than the artificial organization substitute, foreign and dissimilar material, no immune rejection, and its origin is widespread, based on convenience, low cost, in order to inject way fat grafts filling, become a plastic surgery area of the commonly used method to improve appearance. Often used to fill in facial deformity, treatment of facial atrophy and other diseases. In aesthetic surgery, to the forehead, nose, cheeks, temporal anti-aging treatment areas such as the root and forehead wrinkles after filling, won the facial young; Still can do self fat breast enhancement, abundant buttocks, and so on. But the low fat pellet transplant survival rate and high absorption rate problem, still affects fat grafts, in clinical extensively and the main problems of the application. To obtain and process the fat cell is the effect of fat cell viabitity key factors, which has been studied extensively.At present, the fat grafts technology, treatment and purification methods of fat cell is not consistent:some scholars suggest standing for a long time, eliminate impurities such as precipitation; And more scholars have suggested that the method of centrifugal separation impurities, because can improve the efficiency of centrifugal purification.which the size of the centrifugation forces is appropriate, it is not clear, the500RPM,1000RPM,1500RPM,3000RPM,6000RPM all someone used. So the author aimed to investigate the effects of centrifugation force on fat cell viabitity, the centrifugation forces is divided into500RPM,1000RPM,3000RPM,and centrifugation time for3minutes in basic experimental and clinical practice, in order to find the appropriate centrifugation forces for the best way to obtain the purified autologous processed lipoaspirate cells in clinical practice and perfect the current fat grafts technology.The experiment was designed to divide into three parts. The first part, through the different centrifugation forces,the ASCs (adipose derived stromal cells ASCs, or say fat mesenchymal stem cells, BMSCs) content is different from fat tissue, the measure ASCs content of500RPM separation3minutes and1000RPM separation3minutes are more than3000RPM separation3minutes, P<0.01, with a statistical significance. And the ASCs content in500RPM separation3minutes are similar to1000RPM separation3minutes,P>0.05, no statistical significance. But500RPM separation3minutes was found that purity is not enough, contains many impurities such as red blood cells, and1000RPM high purity. The second part, three centrifugation forces from the fat cells histopathologic study, was found that500RPM centrifugal after3minutes of cell damage minimum, and3000RPM, centrifugal after3minutes of injury is the largest, and1000RPM separation after3minutes, cells have slightly damaged. The third part, from clinical application, the self fat breast augmentation and facial of patients filled, respectively, after transplantation in three months, six months, nine months follow-up, and through the patients, doctors and the third party carried on the questionnaire survey, and concludes that the1000turn/separation3minutes is the best effect and the highest satisfaction after autologous adipose tissue of breast augmentation and facial depression filling.To sum up, in view of the cells from centrifugal purity, the content of ASCs,cell damage minimization, clinical application,et al.several aspects of contrast, draw liposuction won adipose tissue, through in the1000RPM speed, centrifugal3minutes after the clinical application, best effect. So as to guide clinical doctors in the fat cell transplantation, choose appropriate centrifugation forces purification fat, so as to improve the survival rate of fat cells, reduce the absorption rate, to achieve the best effect of transplantation. Let the fat grafts in clinical extensively and application.
