Charpter1Expression of A2M in the serum of AR patientsObject To test the expression levels of A2M in the serum of AR patients. Exploring the correlation of A2M expression with AR pathogenesis.Methods Using cELISA technique to detect the levels of A2M in the serum of patients from two groups:healthy group and AR patients group,each group has20people. The correlation between the A2M expression and AR pathogenesis was analyzed statistically.Results Expression of A2M was significantly upregulated in AR patients compared to the healthy volunteers.Conclusion The expression of A2M was significantly increased in serum of patients with AR, prompting to us to speculate that A2M may play an important role in the pathogenesis of AR. Finally to further study the potential role of A2M、 STAT3、IL22、 IL17and A2M-Stat3-TH17pathway in AR pathogenesisMethod Using immunofluorescence method and double immunoflu-orescence labeling to check A2M、STAT3、 IL22、 IL17expression and A2M+STAT3、 STAT3+IL22、 STAT3+IL17co-localization in6non-inflammatory nasal mucosa specimens and6AR nasal mucosa specimens.Results Each nasal speciman has A2M、 Stat3、 IL22、 IL17positive cells and A2M+Stat3、 STAT3+IL22、 STAT3+IL17co-labeling cells, we have counted the single A2M、 Stat3、 IL22、 IL17labeling cells and A2M+Stat3、 STAT3+IL22、 STAT3+IL17double labeling positive cells. AR group has significantly single labeling cells and co-labeling cells than the normal group.Conclusion Expression of A2M、 Stat3、 IL22、 IL17in the nasal mucosa from AR patients was significantly higher than the control group, prompting that they may be involved in the pathogenesis of AR, Further double immunofluorescence labeling experiments show that A2M+Stat3、 STAT3+IL22、 STAT3+IL17double labeling positive cells in AR group was significantly higher than control group, prompting that the A2M-Stat3-TH17pathway may be involved in the pathogenesis of AR. Specific blocking this pathway might be helpful for the treatment and prevention of AR.
