Objective To investigate the role of IL17 signaling in the immunity response of HIV and Penicillium marneffe(PM) co-infected individuals.Methods (1)Immunohistochemical method:determine the expression of CD4+,IL17,TRAF6,NFκB p65 and IL22 in the skin tissue.7 HIV and PM coinfected individuals as Test group Ⅰ; 9 HIV infected dermatitis individuals as Control group 1; 9 PSM with PAS-positive individuals as Control group 2; 9 PSM with PAS-negative individuals as Control group 3;6 healthy person as Control group 4.(2)ELISA:determine the expression of IL17,TRAF3IP2, TRAF3,TRAF6,NFκB p65,TNF-αand IL22 in the serum.16 HIV infected cases as Control group I;32 HIV-positive oral candida cases as Control group II, and 23 cases of them with CD4+T cells<50×109/L as Control group Ⅱa,9 cases with CD4+T cells≥50×109/L as Control group Ⅱb; 16 healthy person as control Ⅲ; 64 HIV and PM co-infected cases as Test group Ⅱ,51 cases of them with CD4+T cells<50×109/L as Test group Ⅱa,13 cases with CD4+T cells≥ 50x 109/L was Test group II b.Results Immunohistochemical staining:(1)The expression of CD4+:the positive rate of CD4+ was (7/7,100%) in Test group I, the difference was no statistical significance compare with Control group 1、2、3 (all P>0.05),but it was statistical significance when compare with Control group 4 (P< 0.05).(2)The expression of IL17:the positive rate of IL17 was (6/7,85.7%) in Test group I, the difference was no statistical significance compare with Control group 1、2、3 (all P>0.05),but it was statistical significance when compare with Control group 4 (P<0.05).(3)The expression of TRAF6:the positive rate of TRAF6 was (4/7,57.1%) in Test group I, the difference was no statistical significance compare with Control group 1 and 3 (all P>0.05),but it was statistical significance when compare with Control group 2 and 4 (all P< 0.05).(4)The expression of NFκB p65:the positive rate of NFκB p65 was (5/7,71.4%) in Test group I, the difference was all statistical significance compare with Control group 1、2、3 and 4 (all P≤0.05).(5)The expression of TNFa:the positive rate of TNFa was (2/7,28.6%) in Test group I, the difference was all no statistical significance compare with Control group 1、2、3 and 4 (all P>0.05).(6)The expression of IL22:the positive rate of IL22 was (7/7,100%) in Test group Ⅰ,and it was the same in the Control group l,but the difference was all no statistical significance compare with Control group 1、2、3 and 4 (all P >0.05). ELISA:(1)The expression of IL17:compared with Control group Ⅲ,the difference was no statistical significance among Control group Ⅰ、Ⅱa and Ⅱb (all P>0.05),but it was statistical significance in Test group II a and Ⅱb(all P<0.05);compared with Control group I, the difference was statistical significance among Control group Ⅱa、Test group Ⅱa and Ⅱb (all P< 0.05),but it was no statistical significance in Control group Ⅱb (P> 0.05);compared with Test group Ⅱa, the difference was no statistical significance among Control group Ⅱ a and II b and Test group Ⅱb (all P> 0.05); compared with Test group Ⅱb, the difference was no statistical significance among Control group Ⅱa and Ⅱb (all P>0.05); the difference was no statistical significance between Control group Ⅱ a and II b (P>0.05),(2)The expression of TRAF3IP2:compared with Control group Ⅲ,the difference was no statistical significance among Control group Ⅰ、Ⅱ a and Ⅱ b and Test group Ⅱa and Ⅱb (all P>0.05);compared with Control group I, the difference was statistical significance among Control group Ⅱa、Ⅱb and Test group II a and Ⅱb (all P <0.05);compared with Test group Ⅱa, the difference was no statistical significance among Control group Ⅱa and II b and Test group Ⅱb (all P> 0.05); compared with Test group Ⅱb, the difference was no statistical significance among Control group II a and II b (all P> 0.05); the difference was no statistical significance between Control group Ⅱa and II b (P>0.05).(3)The expression of TRAF3:compared with Control group Ⅲ,the difference was no statistical significance among Control group Ⅰ、Ⅱa and Ⅱb and Test group II a and Ⅱb (all P>0.05);compared with Control group I, the difference was no statistical significance among Control group Ⅱa、Ⅱb and Test group Ⅱb (all P > 0.05), but it was statistical significance in Test group II a (P< 0.05);compared with Test group II a, the difference was no statistical significance among Control group II a and II b and Test group Ⅱb(all P>0.05); compared with Test group Ⅱb, the difference was no statistical significance among Control group II a and II b (all P>0.05); the difference was no statistical significance between Control group II a and II b (P>0.05).(4)The expression of TRAF6:compared with Control group III,the difference was no statistical significance among Control group Ⅰ、Ⅱ a and Ⅱb and Test group II a and Ⅱb (all P>0.05);compared with Control group I, the difference was no statistical significance among Control group Ⅱa、Ⅱb and Test group Ⅱa and Ⅱb (all P>0.05);compared with Test group II a, the difference was statistical significance among Control group Ⅱ a and Ⅱb (all P<0.05),but it was no statistical significance in Test group Ⅱb (P>0.05); compared with Test group Ⅱb, the difference was statistical significance in Control group Ⅱa (P< 0.05),but it was no statistical significance in Control group Ⅱb (P>0.05); the difference was no statistical significance between Control group II a and Ⅱb (P >0.05).(5)The expression of NFκB p65:compared with Control group Ⅲ,the difference was statistical significance among Control group II a and II b and Test group Ⅱa and Ⅱb (all P<0.05),but it was no statistical significance in Control group ~ (P> 0.05);compared with Control group I, the difference was statistical significance among Control group Ⅱa and II b and Test group Ⅱa and Ⅱb (all P<0.05);compared with Test group Ⅱa, the difference was statistical significance among Control group Ⅱa and Ⅱb (all P<0.05),but it was no statistical significance in Test group Ⅱb (P>0.05); compared with Test group Ⅱb, the difference was no statistical significance in Control group Ⅱa and Ⅱb (P>0.05); the difference was no statistical significance between Control group Ⅱ a and Ⅱb (P>0.05).(6)The expression of TNF-a:compared with Control group Ⅲ,the difference was no statistical significance among Control group Ⅰ、Ⅱa and Ⅱb and Test group Ⅱa and Ⅱb (all P> 0.05);compared with Control group Ⅰ, the difference was no statistical significance among Control group Ⅱa and Ⅱb and Test group Ⅱa and Ⅱb (all P> 0.05);compared with Test group Ⅱa, the difference was statistical significance among Control group Ⅱa、 Ⅱb and Test group Ⅱb (all P>0.05); compared with Test group Ⅱb, the difference was no statistical significance in Control group Ⅱa and Ⅱb (all P> 0.05); the difference was no statistical significance between Control group Ⅱa and Ⅱb (P>0.05).(7)The expression of IL22:compared with Control group Ⅲ,the difference was no statistical significance among Control group II a and Ⅱb and Test group II a and Ⅱb (all P>0.05), but it was statistical significance in Control group I (P<0.05);compared with Control group I, the difference was no statistical significance among Control group II a and II b and Test group Ⅱa and Ⅱb (all P>0.05);compared with Test group Ⅱa, the difference was statistical significance among Control group Ⅱa、Ⅱb and Test group Ⅱb(all P>0.05); compared with Test group II b, the difference was no statistical significance in Control group Ⅱa and Ⅱb(all P>0.05); the difference was no statistical significance between Control group II a and Ⅱb(P>0.05).Conclusion (1)The four infection states that HIV infected、HIV and PM coinfected、HIV and Mycotoruloides coinfected and HIV-negative PM infected, existing some differences among them,all can activate the IL17 signaling pathway, but there may be some excessive suppression which makes function decline.(2)HIV and PM coinfected can activate the IL17 signaling pathway along with function declining in peripheral blood and skin lesion,,but there may be some differences in both tissues.
