Objective: The present study was designed to discussion and analysis ofcombination therapy of Shenqi Yigan Decotion and5-Fu. For two groups ofcombination of Chinese and western medicines, the effect of the expression of TLR4and PCNA,survival conditions in H22tumor-bearing mice.Methods: Hepatoma22tumor bearing mice models were established, then themice (n:80) were divided randomly into four groups:Normal Saline negativegroup(NS),5-Fluorouracil (5-Fu)positive treatment group, Shenqi Yigan Decotiontreatment group and Drug combination therapy group. Then according to the group fordrug treatment, respectively. Then at the time of delivery and daily observation, recordthe different groups tumor models in mice survival and weight change.Drug delivery in15days in a row, at the end of the dosing after24hours, will be put to death all tumormodel mice. Measurement of all the final weight and the tumor weight in mice tumormodel. Calculate the average tumor weight and the inhibiting rate of tumor, analysis ofweight change trend, and carry on the comparison between the groups.Apoptosis factorsTLR4and PCNA were detected by immunohistochemical method.And comparing withthe result of each group of data analysis.Results:The inhibiting rate of tumor were56.8%,6.5%and66.2%in5-Fupositive treatment group,Shenqi Yigan Decotion treatment group and Drug combinationtherapy treatment group. Immunohistochemical method to detect, between groups ofH22 liver cancer cells in mice tumor model of TLR4and PCNA expression in tumortissue.Results showed that the t test analysis: The expression of TLR4and PCNA were increased in three drug groups compared with NS negative group.5-Fu positivetreatment group and Drug combination therapy treatment group both have obviouslyless expression than NS negative group (P<0.05), the difference had statisticalsignificance.But the expression of Shenqi Yigan Decotion treatment group wasincreased little (P>0.05), the difference had no statistically significant. And5-Fupositive treatment group compared with, Shenqi Yigan Decotion treatment group ishiger (P <0.05), Drug combination therapy group is lower (P <0.05), there is statisticalsignificance too. And compared with Shenqi Yigan Decotion treatment group, Drugcombination therapy group is lower (P <0.05), there is statistical significance.According to the weight change, each group the average tumor weight and survival state,four groups were compared.5-Fu treatment group final weight is less than the initialweight in mice tumor model, the rest of the group are weight gain in mice. ShenqiYigan Decotion treatment group have most of the weight gain in mice. The averagetumor weight in mice of NS negative group is the biggest. The rest of the group theaverage tumor weight of mice were lower than the group. The average tumor weight inmice of Drug combination therapy group is the smallest. Survival conditions of5-Fupositive treatment group is the worst, contrarily Drug combination therapy treatmentgroup is groove and weight change is the most stable. Survival conditions of the othertwo groups are similarly.Conclusion: The anti-tumor action of Shenqi Yigan Decotion is not obviously.Compared with unitarily5-Fu positive treatment group, Drug combination therapytreatment can not only increase anti-tumor action on Hepatoma22tumor,down-regulating TLR4and PCNA expression, but also show significant effect onraising survival conditions.
