Objective:To oberve the expression of suppressors of cytokine signaling three(SOCS3)proteion, detect the frequency of help T cell one(Th1) and help T cell two(Th2)andthe bias of Th1/Th2and IgA-produce B cell in peripheral blood T lymphocyte cell ofIgAN patients,and analyze the correlation with clinical and pathological parameters,explore The effect of suppressor of cytokine signalling proteins3(SOCS3) in theRegulation of Th-cell polarization on the immunological mechanism of IgAnephropathy.Methods:25IgAN patients and20healthy normal controls with comparable age andGender distribution were researched. All patients were pathologic diagnosed primaryIgA nephropathies excluding secondary reasons.None of the patients have beentreated with glucocorticoid or immunosuppressants in one month.The PBMC of IgAN patients and normal controls were separated by Ficollmethod.The SOCS3protin lelvel,The frequency of Th1,Th2and IgA-produce B cellsin peripheral blood of IgAN patients and heathy comtrols were detected byintracellular cytokine detection method with flow-cytometry and western blot(WB),and the ratio of Thl/Th2was calculated.The concentration of IgA1and its capabilityof combinging with VVL were examined by ELISA.Result:1. The levels of Thl in IgAN decreased in IgAN compared to those of healthycontrols,but no significant difference.While The percentage of Th2were significantlyincreased in IgAN patients compared to healthy controls,respectively were2.56±0.67%VS1.64±0.51%.The percentage of IgA-produce B cell increased in IgANpatients compared to the controls,respectively were12.98±4.73%VS1.81±1.47%.Accordingly the ratio of Thl/Th2reduced. The frequency of Th2waspositively correlated with serum IgA level(r=0.64, P <0.05). No significantcorrelation between TTh2and clinical or pathological parameters was detected. 2. Compared to The healthy controls The expression of SOCS3protein inperipheral blood mononuclear(PBMC) was significantly increased in IgAN patients(P <0.05),and was positively correlated with the percentage of Th2(r=0.445, P<0.05).3. The concentration of IgA1in serum of IgAN patients were significantlyhigher than those of the healthy controls,respectly were1.92±0.67VS1.74±1.02mg/ml(P <0.05). Compared to the normal controls,the capacity to combine with VVLof IgA1from IgAN patients increased, respectly were4.67±0.59VS2.94±0.96AU/mg.Conclusion:(1) The bias of Th1/Th2was found towards Th2in IgAN patients,Th2couldcontribute to developpment of IgAN by producing exceed IgA1with aberrantglycosylation.(2) The expression of SOCS3protein in PBMC was significantly increased inIgAN patients compared to the normal controls,SOCS3may participate in theimmunepathogenesis of IgAN by regulating the polarization of Th cell.
