Background:At present, the hazards of silicosis in china are still the most severe,and the people contacting silica dust, the new cases of silicosis and the patients ofsilicosis are the highest in the world. The mechanism of silicosis formation hasn’tbeen clear up to now. Though each of them has their own basis of theory, but it is notentirely reasonable to explain its pathogenicity process. In recent years, TGF-β bymost experts,recognized the critical cytokine for the formation and development ofpulmonary fibrosis. TGF-β showed common fibrogenic role in a variety of fiberproliferative diseases. And the TGF-beta1the most important subtype molecular ofTGF-beta ‘s family. Smad7protein is a newly discovered protein, is downstreamprotein of TGFβ-depended Smads signal pathway.Abnormal expression of theSmad7protein play a key role in the development of pulmonary fibrosis.Many studieshave shown that TGF-β1and Smad7play an important role in the pathogenesis ofpulmonary fibrosis.In this experiment,the expressions of both TGF-β1mRNA andSmad7mRNA of the rabbits lung tissue were monitored in experimental group andthe control groups, which explore its possible role in the pathogenesis of silicosisfibrosis.Objective:To investigate the effects of cordyceps sineses in rabbit silicoticfibrosis model and succeed in creating rabbit silicotic model.Methods:Thirty health male rabbits were randomly divided into3groups:1、control of blank group(BP),2、control of silicotic model group(CP),3、experimentalgroup(EP).The rabbits of CP and EP groups were perfused with the mix liquid ofsilicon dioxide (120mg/kg) endotracheally,and BP group with normal saline insteadunder the same conditions.All the rabbits were sacrificed after30days,CalculatedLung/Body coefficient by weighting the lung weight and the body weight ofrabbits,The right lung tissue was stained with hematoxylin-eosin(HE).Content ofHydroxyproline were measured by alkaline hydrolysis,The levels of the expression ofTGFβ-1and Smad7mRNA of rabbits lung sections were measured by Real-timePCR. Results:We observed the normal lung tissue in blank group by HE, and thefibrosis and silicotic nodes in the control of silicotic model group and experimentalgroup by HE. The Lung/Body coefficient and the content of Hydroxyproline ofrabbits lung tissue were apparently differences in the three groups. Compared withthe rabbits of BP, TGF-β1mRNA of rabbits lung sections were significantly higherand Smad7mRNA of rabbits lung sections were significantly lower in EP; Comparedwith the rabbits of CP, TGF-β1mRNA of rabbits lung sections were significantlylower and Smad7mRNA of rabbits lung sections were significantly higher in EP.Conclusion:The cordyceps sinesis may play an important effect in the formationof rabbits silicotic fibrosis, and there may be reduces the expression of TGFβ-1mRNA and updowns the level of Smad7by TGFβ-depended Smads signalpathway,which postpone the formation of in rabbit silicotic fibrosis。...
