Objective:1. To detect the activity of SOD and the content of MDA in mouse skin andliver tissue, and observe the changes of weight, behavior and pathomorphology of skinmelanocytes in mice. To establish the evaluation norms of melasma model mouses withliver-qi depression systematically and scientifically.2. To explore the efficacy ofxiaobanling to melasma model mouses with liver-qi depression, and explain themechanism preliminarily. To provide data to support xiaobanling into clinical research.Methods:1. After open field test, and tail suspension test, we chose the Kunming mice ofhealth and female into test, then made model after partially ultraviolet radiation, theinjection of progesterone, and chronically constrained. We evaluated model on changes ofthe activity of SOD and the content of MDA in mouse skin and liver tissue, and thechanges of weight, behavior and pathomorphology of skin melanocytes.2. We chose theappropriate mice into the experiment according to the above-mentioned methods. Theywere divided into normal group, model group, Xiaobanling high-dose group, middle-dosegroup and low-dose group, and positive control group. Then they were given drugintervention after modeling and observed of the changes of these indicators.Results:1. Compared with normal group after molded28d, the weight and total score ofOFT lessened significantly(P<0.01), the immobility time of TST increased significantly(P<0.01); the SOD activity in mouse skin and liver tissue of the model group decreasedsignificantly (P<0.05, P<0.01), MDA was increased significantly(P<0.01, P<0.001). Inskin melanocytes Immunohistochemical assay, no melanocytes was saw in the skin ofnormal mice, but increased significantly in the skin of model mice and had a strong expression. The number of melanocytes, MOD, number density were increasedsignificantly(P<0.001).2. Compared with model group, the macroeconomic indicators ofmice of Xiaobanling high-dose group, middle-dose group, low-dose group and positivecontrol group improved significantly (P<0.05, P<0.01, P<0.001). The SOD activity inmouse skin and liver tissue of the Xiaobanling high-dose group increased significantly(P<0.05), MDA of Xiaobanling middle-dose group reduced significantly(P<0.01). Thenumber of melanocytes in prickle cell layer and the basal cell layer of drug-treated groupreduced significantly, and Melanin-positive expression was significantly reduced.Conclusion: The method of partially ultraviolet radiation+the injection of progesteroneand chronically constrained establishes the chloasma mouse model with Liver-Qidepression successfully; Xiaobanling has a significant role in the treatment of the chloasmamouse model with Liver-Qi depression. The mechanism of action may be related toimprove the oxidation resistance of body’s, inhibit the activity of tyrosinase, improve bloodrheology, and enhance the body’s function of soothing Liver-Qi stagnation.
