Objective: Two treatments were selected in this study—herbs-separated umbilicalmoxibustion combined with needling Sanyinjiao and herbs-separated umbilicalmoxibustion to treat the patients with primary dysmenorrhea. By detecting spectrummetabolites in patients,plasma and urine samples of the two groups before and aftertreatment to identify potential biomarkers and derive metabolic pathways, whereby explorethe likely effective targets of treating the disease by herbs-separated umbilical moxibustioncombined with needling Sanyinjiao and herbs-separated umbilical moxibustion. Comparethe metabolic pathways, the similarities and differences in regulating the uterus of the twogroups.Methods:20patients with primary dysmenorrhea were randomly divided into twogroups,10cases in the group of herbs-separated umbilical moxibustion combined withneedling Sanyinjiao;10cases in the group of herbs-separated umbilical moxibustion. Thenwe dealed with the patients,plasma and urine samples of the two groups before and aftertreatment. According to the Simca-P11.5software package processing data, looked for thecontributing metabolite fractions and searched for related metabolic pathway by HMDB、KEGG.Result: The metabolite spectrum of the two groups had undergone significant changesand the more significant changed metabolites of both groups were steroids, prostaglandins, catecholamine, histamines, neurotransmitters, amino acids and so on. The metabolites ofthe two groups were commonly involved in the body’s steroid hormone synthesis,prostaglandins metabolism, catecholamine metabolism, histamine metabolism,neurotransmitters, amino acid metabolism and so on. In addition, the group ofherbs-separated umbilical moxibustion combined with needling Sanyinjiao was alsoinvolved in the body’s nitric oxide synthesis, oxytocin metabolism, signal transduction andso on.Conclusion: The metabolic mechanisms of herbs-separated umbilical moxibustioncombined with needling Sanyinjiao and herbs-separated umbilical moxibustion on treatingthe patients with primary dysmenorrhea were to adjust patients,abnormal reproductiveendocrine hormone level, to improve their nervous, immune and endocrine network statusto reduce pain and to relieve its accompanying symptoms of primary dysmenorrhea. Thegroup of herbs-separated umbilical moxibustion combined with needling Sanyinjiao wassignificantly better than the group of herbs-separated umbilical moxibustion in relievingdysmenorrhea symptoms. It could be deduced that herbs-separated umbilical moxibustionand acupuncture Sanyinjiao point had a synergistic effect in regulating the uterus.
