Experimental Studies On Protection Of Medicine-separated Moxibustion At "Shenque" On Gastric Mucosal Lesion In Stress Ulcer(SU) Rats

Objective: To explore mechanism of medicine-separated moxibustion at "Shenque'" of preventing experimental stress ulcer by means of ulcer index(UI) , gastric mucosa blood flow(GMBF) gastric mucosa calcitonin-gene-related peptide(CGRP) prostaglandin(PGE2) transforming growth factor(TGF ) and serum tumor necrosis factor(TNF ), thus it could provide theory foundation for effective clinical prophylaxis and treatment of SU; to prove superiority of medicine-separated moxibustion and discuss the action agents of moxibustion though comparing with effect among every group.Methods: 60 rats(30 male rats, 30 female rats) were randomly completely divided into 6 groups: normal group, model group, pure moxibustion at "Shenque" group ( pure moxibustion group ) , medicine-separated moxibustion at "Shenque" group (medicine-separated moxibustion group) , medicine-separated moxibustion at non-point group (non-point group) , warm-up by light at "Shenque" group(warm-up by light group), each group has 10 rats. Each group (rats of normal group and model group were only bound in cage) was respectively pretreated by use of pure moxibustion and medicine-separated moxibustion as well as warm-up by light for 10 days.After 10 days7 pretreatment, the rats(except normal group) were exposed to water restraint stress(WRS) for 12 hours. Then GMBF was measured by LDF Flowmeter, the index of gastric mucosal lesions(UI) were evaluated grossly by means of Guth's methods, content of TGF- , PGE2 CGRP was examinated by radio-immunity method; content of TNF- was examinated by enzyme-linked immunoadsordent assay.Results:l.UI of pure moxibustion at "Shenque" group and medicine-separated moxibustion at "Shenque" group was obvious less than that of medicine-separated moxibustion at non-point group and warm-up by light group (P<0. 01), and the latter two groups have no statistical difference compared with model group respectively ( P>0. 05 ) . 2. Pure moxibustion and medicine-separated moxibustion at "Shenque" as well as warm-up by light group could promote GMBF with significant differences compared with model group and non-point group respectively (P<0. 01), and there were significant differences between moxibustion at "Shenque" groups and warm-up by light group respectively (P<0. 01 or P<0. 05).3.Content of mucosal CGRP and PGE2 in pure moxibustion at "Shenque" group and medicine-separated moxibustion at "Shenque" group both had increased to some degree, it has a clear difference compared with model group, and their activity of upgrading content of CGRP are clearly stronger than moxibustion at non-point group and warm-up by light group (P<0. 01). Medicine-separated moxibustion at "Shenque" had brought about the optimal effect.4. Pure moxibustion and medicine-separated moxibustion at "Shenque" as well as warm-up by light could all promote release and excretion andsynthesis of TGF- with distinct difference compared with model group and non-point group respectively (P<0. 01). And medicine-separated moxib-ustion at "Shenque" had brought about the optimal effect, then pure moxib-ustion at "Shenque"group was the next.5. Pure moxibustion and medicine-separated moxibustion at "Shenque" as well as warm-up by light could all decrease serum content of TNF- with significant difference compared with model group and non-point group respectively (P<0. 01). And medicine-separated moxibustion at "Shenque" had brought about the optima! effect, then pure moxibustion at "Shenque"group was the next.Conclusions:i The experimental study proved that medicine-separated moxibustion at "Shenque" point have protective effect on gastric mucosal damage in SU rats to a great extent.2. The possible mechanism was in that improvement of GMBF and acceleration of release and excretion and synthesis of CGRP PGE2 TGF- as well as inhibition of release of serum TNF- .3 The protective effects of medicine-separated moxibustion at "Shenque" were superior to pure moxibustion at "Shenque" point and non-point group and warm-up by light group ,which could confirm the superiority of medicine-separat...
